钙代谢相关基因与单纯性肥胖症关系的研究 被引量:2

摘要 [目的]探讨高脂饮食诱导下肥胖易感(obesity-prone,OP)与肥胖抗性(obesity-resistant,OR)大鼠在体重、脂肪湿重及白色脂肪组织(white adipose tissue,WAT)中钙代谢相关基因脂肪酸合成酶(fatty acid synthase,FAS)、维生素D受体(vitamin D receptor,VDR)、解偶联蛋白2(uncoupling protein 2,UCP2)的表达差异.[方法]72只SD大鼠, 随机选取12只为标准对照组,基础饲料饲养7周,余60只基础饲料饲养1周后,按体重增值分为OP组、OR组和高脂组, 分组后改用高脂饲料继续饲养6周,观察各组大鼠体重变化、脂肪湿重等;用RT-PCR技术分析WAT中FAS、VDR、 UCP2基因mRNA表达水平.[结果]①OP组体重增值、脂肪湿重均高于OR组(P<0.05);②OP组FAS基因mRNA 表达水平高于OR组(P<0.05);OR组UCP2基因mRNA表达水平则显著高于OP组、高脂组和标准组(P<0.001),后三组问差异无显著性;OP、OR、高脂组三组间的VDR mRNA无差异,但均低于标准组(P<0.05)。[结论]OP与OR 组间存在的生理学差异,与OP组FAS表达增强、UCP2表达减弱导致产热减少密切相关,与VDR的参与可能有关。 [Objective] To explore the differences ot partial physiologic indxes iincludiing weight ,the wet wcight of pad and genetic expression of calcium metabolic relative genes as fatty acid synthase(FAS), vitamin D receptor(VDR) and uncoupling protein 2 (UCP2) mRNA in white adipose tissue(WAT) between obesity-prone(OP) and obesity-resistant (OR) rats on a high-fat diet. [Methods] After 1 week of free access to a basic diet, 12 rats were selected randomly as standard control group and continue fed on basic diet, the 60 rats remains were separated on the hasis of 1 week body weight percentage gained to obesity-prone(OP,≥75) or obesity-resistant ( OR,≤25) groups, and were changed into feeding the high-fat diet for another 6 weeks. The body weight and wet weight of fat pad were measured at the end of killing rat. And the expression levels of calcium metabolic relative genes as FAS, VDR and UCP2 mRNA in WAT were detected by RT-PCR technique. [Results] ①In OP rats, the speed of body weight gain, the wet weight of fat pad were higher than those in OR rats(P〈0.05). ②There was a higher level of FAS mRNA expression in OP rats compared with those in OR rats(P〈0.05) ;the expression level of UCP2 in OR rats was significant higher than those in the others(P〈0.001 ) ;there was no significant differences of VDR mRNA in OP, OR and high-fat rats, but the differences between the three comparing with standard control rats were existed(P〈0.05). [Conelusions] The reasons of physiologic differences between OP and OR rats are as follow;OP rats have a higher expression of FAS mRNA ,a less expression of UCP2 mRNA and a less heat pro duction than those in OR rats. And it shows that the relationship with VDR may he involved to the differences.
