急性暴露尼古丁对1月龄大鼠丘脑腹后内侧核内Caspase-3的表达及凋亡的影响 被引量:3

摘要 目的研究急性暴露尼古丁(n icotine)对1月龄大鼠丘脑腹后内侧核内caspase-3表达及凋亡的影响。方法腹腔注射尼古丁(1mg/kg)建立急性暴露尼古丁的动物模型,用免疫组织化学方法检测模型鼠丘脑腹后内侧核caspase-3的表达,用原位末端标记法(Tunel法)检测凋亡细胞。结果在模型鼠丘脑腹后内侧核,神经细胞caspase-3表达高于对照组。Tunel法证明尼古丁暴露3天组丘脑腹后内侧核凋亡细胞百分率与对照组无明显差异,尼古丁暴露6天组丘脑腹后内侧核凋亡细胞百分率明显多于对照组。结论急性暴露尼古丁使1月龄大鼠丘脑腹后内侧核内caspase-3表达增加,促进该核团内的神经细胞凋亡。 Objective To study the effect of acute nicotine expesure on expression of Caspase-3 and apoptosis of ventropesterior medial nucleus (VPM) of thalamus in 30 days aged rat. Method The animal model of rats was expesed to nicotine intraperitoneally. The Caspase-3 expression was detected with immunohistochemistry and apoptotic cells were detected by Tunnel method. Result After nicotine was administrated for 3 days and 6 days respectively, Caspase-3 expression in VPM is higher in nicotine exposed group than in control group. After 3 days nicotine exposure, the percentage of the apoptotic cells in VPM in nicotine exposure group was similar to that of the control group but the percentage of the apoptotic ceils in nicotine exposure group after 6 days nicotine exposure were more than that in the control group. Conclusion Acute nicotine exposure increased Caspase-3 expression and accelerated neuron apoptosis in VPM of the 30 days aged mrs.
