BCEF0083对慢性应激抑郁大鼠海马脑源性神经营养因子含量的影响

Impact of BCEF0083 on content of hippocampal brain-derived neurotrophic factor in rats with chronic stress-induced depression

摘要 目的:观察一种白僵菌代谢产物BCEF0083对慢性应激抑郁模型大鼠糖水消耗量和海马脑源性神经营养因子含量的影响。方法:实验于2003-10在安徽医科大学药理学教研室完成。①分组:选用雄性SD大鼠48只,随机分为6组:对照组、模型组、吗氯贝胺20mg/kg组、BCEF0083100,50,25mg/kg组,每组8只。②模型制备:采用慢性不可预见性应激法造成大鼠抑郁模型。③给药:用药组灌胃给药,1次/d,共21d,模型组及对照组灌胃蒸馏水,吗氯贝胺组灌胃剂量为20mg/kg、BCEF0083大剂量组灌胃剂量为100mg/kg,BCEF0083中剂量组灌胃剂量为50mg/kg,BCEF0083小剂量组灌胃剂量为25mg/kg。④用糖水消耗量法观测大鼠行为,用免疫组织化学测定海马脑源性神经营养因子的含量。结果:参加实验48只大鼠,全部进入结果分析。①在糖水消耗实验中:与对照组比较,模型组大鼠糖水消耗量降低(P<0.01);与模型组比较,BCEF0083100mg/kg组、BCEF008350mg/kg组、BCEF008325mg/kg和吗氯贝胺20mg/kg组均能增加慢性应激大鼠糖水消耗量(P<0.01或P<0.05)。②在BCEF0083对慢性应激大鼠海马脑源性神经营养因子含量的影响实验中:与对照组比较,模型组大鼠海马CA1,CA3,齿状回区的脑源性神经营养因子阳性神经元平均吸光度减小(P均<0.01);与模型组比较,BCEF008325mg/kg组,50mg/kg组和吗氯贝胺20mg/kg组均能够不同程度分别增加慢性应激大鼠海马CA1,CA3,齿状回区的脑源性神经营养因子阳性神经元平均吸光度(P均<0.01);与对照组比较,模型组大鼠海马,CA3,齿状回区的脑源性神经营养因子阳性神经元面积百分比减小(P均<0.01);与模型组比较,BCEF008325mg/kg,50mg/kg均能不同程度分别增加慢性应激大鼠海马CA1,CA3,齿状回区的脑源性神经营养因子阳性神经元面积百分比(P<0.01或P<0.05)。结论:BCEF0083的抗抑郁作用可能与海马脑源性神经营养因子含量有关。 AIM: To observe the effects of BCEFO083, metabolite of botrytis bassiana,on sucrose consumption and content of hippocampus brain-derived neurotrophic factor in rat models with chronic stress-lnduced depression. METHODS: The experiment was performed at Department of Pharmacology, Anhui Medical University in October 2003. ①Grouping: Totally 48 male SD rats were selected and randomly divided into six groups: control group, model group, moclobemide 20 mg/kg group, BCEFO083 100, 50, 25 mg/kg groups with 8 rats in each group. ②Model preparation: The depression model rat was produced by chronic unpredictable stress. ③Administration: The rats in the drug group were treated with drug by gastric perfusion, once a day for 21 days. Those in the model group and control group received distilled water by gastric perfusion, 20 mg/kg in moclobemide group, 100 mg/kg in BCEFO083 large dose group, 50 mg/kg in BCEFO083 middle dose group, 25 mg/kg in BCEF0083 small dose group. ④Behavior of rats was determined with sucrose consumption. The content of hippocampus brain-derived neurotrophic factor was determined with immunohistochemical method. RESULTS: A total of 48 experimental rats were involved in the result analysis.①Sucrose consumption trial: Compared with the control group, the sucrose consumption reduced in the model group (P 〈 0.01). Compared with the model group, the sucrose consumption in rats with chronic stress-induced depression increased in the BCEF0083 100 mg/kg group, BCEF0083 50 mg/kg group, BCEF0083 25 mg/kg and moclobemide 20 mg/kg group (P 〈 0.01 or P 〈 0.05).②Effect of BCEFO083 on content of hippocampus brain-derived neurotrophic factor in rats with chronic stress-induced depression: Compared with the control group, mean absorbance (A) decreased in positive neuron of brain-derived neurotrophic factor in hippocampal CA1,CA3 and dentate gyrus regions in model group (P 〈 0.01). Compared with the model group, mean A could increase to certain extent in positive neuron of brain-derived neurotrophic factor in hippocampal CA1, CA3 and dentate gyrus regions of rats with chronic stress-induced depression in the BCEF0083 25 mg/kg group, BCEF0083 50 mg/kg and moclobemide 20 mg/kg group, respectively (P 〈 0.01). Compared with the control group, percentage of positive neuron area of brain-derived neurotrophic factor decreased in hippocampal CA3 and dentate gyrus regions of rats in the model group (P 〈 0.01). Compared with the model group, percentage of positive neuron area of brain-derived neurotrophic factor could increase in hippocampal CA1, CA3 and dentate gyrus regions of rats in the BCEF0083 25 mg/kg and 50 mg/kg groups to certain extent, respectively (P 〈 0.01 or P 〈 0.05). CONCLUSION: The anti-depression effect of BCEFO083 may be associated with the content of hippocampus brain-derived neurotrophie faetor.
