摘要
采用CTAB法,对红掌的叶、叶柄、肉穗花序、佛焰苞、根等5个部位进行基因组DNA提取,利用紫外分光光度法、琼脂糖凝胶电泳法和RAPD扩增法检测DNA的质量,结果表明,CTAB法适合红掌根和叶的基因组DNA提取,没有蛋白质、酚及色素等杂质的污染,佛焰苞的DNA中有少量蛋白质污染,叶柄DNA中含有较多的蛋白质、酚等杂质,而从肉穗花序中没有提取到基因组DNA。
Genomic DNAs from lamina, leaf stalk, mot, spathe, and spadix of Anthurium andraeanum were extracted by the CTAB method. The qualities of DNA were detected by ultraviolet spectrophotometry, agarose gel electrophoresis and RAPD amplification, and the results showed that the CTAB method was adaptive to the genomic DNA extraction of root and lamina because there was no contamination of protein, phenol etc. The genomic DNA from spathe had a few protein contaminations and the genomic DNA from the leaf stalk of A. andraeanum mixed with a lot of protein, phenol and other contaminations, but there was no genomic DNA isolated from spadix by the CTAB method.
