摘要
目的分析尿多酸肽注射液和外周血单核巨噬细胞在其作用下产生的细胞因子两者浓度之间的关系,探讨尿多酸肽对细胞免疫功能的影响。方法选择有明确病理诊断的肿瘤患者和与之年龄、性别相匹配的健康查体者各30例,抽取其静脉血,用梯度密度离心法分离其外周血单个核细胞(PBMCs),用10%胎牛血清1640培养液将其浓度调成1×106/m l,分别加入不同浓度的尿多酸肽注射液,在37℃的二氧化碳培养箱(浓度5%)中培养72 h后,收集细胞悬液-70℃冻存,经反复冻融、离心收集上清液,用酶联免疫吸附法(ELISA)测定上清液中肿瘤坏死因子-α(TNF-α)的含量。结果肿瘤患者PBMCs在加入不同浓度的尿多酸肽培养72 h后上清液中TNF-α的含量分别为(383±133)pg/m l,(377±143)pg/m l,(375±138)pg/m l和(367±149)pg/m l,健康人则分别为(474±161)pg/m l,(469±175)pg/m l,(481±163)pg/m l和(461±183)pg/m l。在相同尿多酸肽浓度下,肿瘤组的TNF-α含量低于健康对照组(P<0.05);但两组的TNF-α含量不随培养液中尿多酸肽浓度的变化而发生明显变化。结论肿瘤患者PBMCs在相同条件下产生的TNF-α低于健康人;尿多酸肽对单核巨噬细胞在体外条件下分泌细胞因子的功能可能无明显影响。
Objective To investigate the effect of uroacitides injection on cell-mediated immunity function by measuring the concentration of eytokines in the supernant collected from the macrophage cells of the patients and the healthy after cultivation . Methods The vein blood from thirty patients and healthy individuals was requried and then the peripheral blood mononuclear cells(PBMCs) was separated with density gradient centrifugation. The PBMCs were plated at a density of 106 cells/ml in RPMI 1640 and treated with uroacitides injection of different concentrations and then incubated at 37℃ in a humidified incubator (5% CO2 ) for 72 hours. The PBMCs were collected and kept at -70℃. After freeze thawing,the cells were centrifuged and the supernatant was gathered. The tumor necrosis factor-α (TNF-α) concentrations in supernatant were determined with ELISA. Results After 72 hours treated with uroacitides injection of different concentrations, the TNF-α concentrations were 383 ±133pg/ml, 377± 143pg/ml, 375 ±138pg/ml and 367± 149pg/ml in different patients' groups respectively,while those of the healthy groups were 474 ±161pg/ml,469±175pg/ml,481±163 pg/ml and 461 ±183pg/ml respectively. At the same concentration of uroacirides injection the TNF-α level of patient group was significantly lower than the healthy group( P 〈0.05 ). But there was nearly no significant difference in the TNF-α levels with the increased concentrations of uroacitides injection (P 〉 0.05 ). Conclusions The TNF-α prodrsced by PBMCS in patients was lower than that of healthy individuals. The results of TNF-α measurement gave a clue that uroacitides injection may have no effect on cytokine secretion of macrophage cells in vitro.
