KAI1复制缺陷型腺病毒载体抑制人胰腺癌细胞迁移实验研究 被引量:4

摘要 目的构建携带人肿瘤转移抑制基因KAI1复制缺陷型腺病毒载体并初步研究其对体外肿瘤细胞迁移的影响。方法用Ad-EasyTM系统大肠杆菌中同源重组的方法,构建Ad-KAI1腺病毒载体,并在293细胞中包装、扩增和纯化,用其感染人胰腺癌细胞并检测KAI1的表达。结果构建的Ad-KAI1可以高效感染人胰腺癌细胞系PANC1,并检测到感染Ad-KAI1的PANC1细胞中KAI1的高表达;同时观察到Ad-KAI1可以抑制胰腺癌细胞系PANC1的迁移。结论携带人肿瘤转移抑制基因KAI1复制缺陷型腺病毒载体可抑制人胰腺癌细胞转移,为KAI1抗胰腺癌转移基因治疗提供依据。 Objective To construct a replication-deficient recombinant adenovirus vector expressing human anti-metastasis gene KAI1 and investigate its migration suppressive effect in vitro. Methods Ad-EasyTM System was used to construct Ad-KAI1 by recombination in E. coll. The virus was packed in 293 cells, identified by PCR and Western blot, amplified effectively and purified by Adeno-XTM Virus Purification Kit. The expression of KAI1 in human pancreatic carcinoma Cells (PANCl) was examined after Ad-KAI1 infection. Results KAI1 protein was highly expressed in PANCl after Ad-KAI1 infection and significantly inhibited the migration ability of tumor cells. Conclusions Successful construction of Ad-KAI1 is of significance in the study of anti-metastasis of pancreatic carcinoma and would prove to be a potential vector for cancer gene therapy.
