摘要
目的探讨首灵健脑胶囊对老年性痴呆小鼠学习记忆及脑内微量元素的影响。方法腹腔注射AlCl3复制慢性铝中毒老年痴呆模型,同时灌胃给予相应药物。给药结束后测试学习记忆成绩、脑系数和脑中微量元素的含量。结果跳台实验显示模型组较正常对照组的错误反应次数明显增多,测试期触电潜伏期明显缩短;给予首灵健脑胶囊或双氢麦角碱(喜得镇)的AlCl3小鼠与模型组比较,训练期和测试期跳台错误反应次数明显减少(P<0.01),测试期触电潜伏期明显延长(P<0.01)。造模后小鼠的脑系数低于正常对照组(P<0.05或P<0.01);中、高剂量的首灵健脑胶囊和喜得镇在使用后,可显著降低脑系数(P<0.05或P<0.01)。模型组小鼠脑中的微量元素Al、Zn、Mn、Ca、Cu、Fe与正常对照组比较,均有统计学意义(P<0.01);用药后,高剂量的首灵健脑胶囊和喜得镇可影响微量元素的含量(P<0.01或P<0.05),中剂量的首灵健脑胶囊可显著影响部分微量元素的含量(P<0.01或P<0.05)。结论首灵健脑胶囊具有明显拮抗铝的神经毒性作用,通过维持脑内微量元素平衡发挥防治老年性痴呆的作用。
Objection To investigate the effect of Shoulinjiannao Capsule on learning memory ability and brains microelements in mice with Alzheimer＇s Disease. Methods AICl3 was abdominal injection to copy chronic aluminum intoxication senile dementia model. After experiment was completed, the learning memory abilities, brain coefficients and microelements in mice brains were measured. Results Jumping stage experiment showed that mistake reaction frequencies in model group was obviously increase than that in normal control group. Electric shock latency periods in model group were obviously shorter than that in normal control group. Jumping stage mistake reaction frequencies in train and test periods for AICl3 mice treated by Shoulinjiannao Capsule or Hydergine were reduced markedly compared with model group（P〈 0.01 ）, and test stage electric shock latency periods were markedly lengthen （P〈 0.01）. Brain coefficients were lower in model group than that in normal control group（ P 〈0.05 or P 〈0.01）. Brain coefficients were significantly lower following the treatment of Hydergine and middle and high doses Shoulinjiannao Capsule（P 〈 0.05 or P 〈 0.01 ）. Contents of Al, Zn, Mn Ca, Cu and Fe in mice brains were significantly different（P〈0.05 or P〈 0.01）. Conclusion Shoulinjiannao Capsule could obviously antagonize aluminous neurotoxic effect by maintained microelements balance.
