摘要
采用超声波浸提法从青蒿中提取青蒿多糖,用硫酸-苯酚法显色,在490nm处运用分光光度计测其含量;并分别利用单因素和正交实验确定了此方法的最佳条件,即单因素法:温度60℃,超声功率80W,固液比1:30,时间30min;各种影响因素影响力的大小为:功率>温度>固液比>时间。正交实验最佳条件:温度70℃,功率90W,固液比1:50,时间40min,为提取青蒿多糖的开发提供了可靠的实验依据。
Ultrasonic-assisted extraction is adopted to extract polysaccharide from artenisia annua L., using sulphuric acid-phenol method and the content of polysaccharide is determined by the spectrophotometer at 490 nm. The optimal conditions are determined with the single factor and orthogonal design, namely, temperature of 60 ℃, power of 80 W and the ratio of solid to liquid 1:30, time of 30 min. The order of different factors are as follows: power 〉temperature 〉solid-liquid ratio〉time. The optimum condition of orthogonal experimental design are: temperature of 70 ℃, power of 90 W , solid-liquid ratio of 1:50 and time of 40 min, which provide an important method for industrial extraction of polysaccharide of artenisia annua L.
