丹参的两种主要成分对培养内皮祖细胞药理作用研究 被引量:16

摘要 目的:观察丹参的两种主要成分丹参素和丹参酮IIA对体外培养内皮祖细胞(EPC)数量和功能的影响。方法:密度梯度离心法获取外周血单个核细胞,培养7天后,收集贴壁细胞并分别加入丹参素5mg/L、10 mg/L及20mg/L,丹参酮IIA0.05mg/L、0.2mg/L及0.8mg/L干预72h。荧光显微镜、流式细胞仪及免疫组化法鉴定EPC,分别观察EPC的数量、增殖、迁移及黏附能力。结果:与对照组相比,丹参素干预组促进外周血EPC扩增,并显著改善外周血EPC的黏附、迁移及增殖能力,丹参酮IIA组对内皮祖细胞数量和功能无明显影响。结论:在丹参的两种主要成分中,丹参素可增加培养EPC的数量并改善其功能,丹参酮IIA则无明显影响。 Objective: To investigate effects of two ingredients of Danshen on number and activity of endothelial progenitor cell in vitro. Methods : Total mononuclear cells(MNCs) were isolated from peripheral blood by Ficoll density gradient centrifugation, and culture in vitro. After 7 days of culture, attached cells were stimulated with Danshensu(5,10and 20mg/L )and DanshinoneⅡA ( 0.05,0.2 and 0.8mg/L) for 72h. The cells were identified by immunohistocbemistry and flow cytometry and tested the ability to intake the acLDL. EPC proliferation and migration were assayed with MTT assay,modified Boyden chamber assay. EPCs adhesion assay was performed by replafing cells on fibronectin -coated dishes,and then counting adherent cells. Results :Dansbensu can increased the number of EPC and promotes EPC proliferative, migratory, adhesive capacity but DanshinoneⅡA not. Conclusion : Danshensu can promote EPC augmentation and enhance its functional activity.
