原花青素对脑缺血后抑凋亡基因蛋白Bcl-2和促凋亡基因蛋白Bax的影响 被引量:15

摘要 目的:观察原花青素对局灶性脑缺血大鼠大脑皮质抑凋亡基因蛋白Bcl-2和促凋亡基因蛋白Bax表达的影响,并分析该影响是否存在剂量依赖性。方法:实验于2004-11/2006-01在锦州医学院药理实验室和免疫组化实验室完成。①选用健康成年SD大鼠30只,雌雄各半,体质量200~250g。②按随机抽签法将大鼠分为5组:假手术组,模型组,原花青素低、中、高剂量组,每组6只。除假手术组不插入线栓外,余下各组均以线栓法阻塞大鼠右侧大脑中动脉,制成局灶性脑缺血模型。以大鼠即刻出现右侧Horner征为造模成功。原花青素低、中、高剂量组:于术前2周分别按100,200,300mg/kg剂量灌胃原花青素混悬液(原花青素粉剂,由天津尖峰天然产物研究开发有限公司提供,批号:20040719),1次/d,共2周,并于术前1h再灌胃1次。假手术组和模型组:于术前2周灌胃等量生理盐水。③术后24h用免疫组化SABC法染色统计大脑皮质每高倍镜视野Bcl-2和Bax表达阳性细胞数。④多组间计量资料差异比较采用单因素方差分析检验,两组间比较采用LSD检验。结果:大鼠30只均进入结果分析。①大鼠大脑皮质Bcl-2阳性细胞数:原花青素低、中剂量组分别为(34.33±8.16)和(31.44±7.09)个,比模型组[(15.72±5.03)个]分别增加了118%,100%,差异明显(P<0.01);原花青素高剂量组与模型组相近[(16.33±5.48)个,P>0.05];原花青素低、中剂量组间差异不明显(P>0.05)。②大鼠大脑皮质Bax阳性细胞数:原花青素低、中剂量组分别为(11.83±4.57)和(12.56±4.31)个,明显少于模型组[(20.5±4.40)个,P<0.01];原花青素低、中剂量组间差异不明显(P>0.05);原花青素高剂量组与模型组相近(P>0.05)。结论:中、小剂量(100,200mg/kg)原花青素可对局灶缺血性脑损伤起到预防性保护作用,其保护作用发生可能与提高Bcl-2表达、抑止Bax表达有关。 AIM: To study effect of procyanidins on expression of Bcl-2 and Bax in rats with focal cerebral ischemic injury and analyze whether the effect is dose-dependent or not. METHODS: The experiment was performed at the Pharmacological Laboratory and Immunohistochemical Laboratory, Jinzhou Medical College from November 2004 to January 2006. ①Thirty healthy adult mate SD mrs, half male and half female rats with the body mass of 200-250 g. ②The rats were randomly divided into the sham operation group, model group and low, middle and high procyanidins groups with 6 rats in each group. The rats were established into focal cerebral ischemic model with middle cerebral artery occlusion (MCAO) except those in the sham operation group. It was succeeded if there was Homer symptom at right side of rats immediately. The rats in the low, middle and high procyanidin groups received 100, 200, 300 mg/kg procyanidin 2 weeks before operation (procyanidin powder, provided by Tianjin Jianfeng Natural Product Research and Development Co., Ltd., No. 20040719), once a day for 14 days. One hour before operation reperfusion was conducted once. The rats in the sham operation group and the model group had been given the equal capacity of saline once 2 weeks before operation. ③The expressions of Bcl-2 and Bax positive cells in cortex of parietal lobe were studied by immunohistochemical SABC method under high power lens 24 hours after MCAO. ④Difference comparison of measurement data among.groups were determined with single factor variance analysis. Comparison between two groups was performed with LSD test. RESULTS: Totally 30 rats were involved in the result analysis.①Number of Bcl-2 positive cells in cerebral cortex of rats: The numbers of Bcl-2 in procyanidins groups with 100 mg/kg and 200 mg/kg were (34.33±8.16) and (31.44±7.09), respectively, increased 118% and 100% as compared with that in the model group (15.72±5.03 ). The difference was significant (P 〈 0.01). It was similar between the high procyanidin group and model group [ (16.33±5.48),P 〉 0.05]. There was insignificant difference between low and middle procyanidin group (P 〉 0.05). ②Number of Bax positive cells in cerebral cortex of rats: It was (11.83±4.57) and (12.56±4.31) in the low and middle procyanidin groups, respectively, which was significandy less than that in the model group [(20.5±4.40) ,P 〈 0;01]. The difference was insignificant between the low and middle procyanidin groups (P 〉 0.05). It was similar between the high procyanidin group and model group (P 〉 0.05). CONCLUSION: Procyanidin in low and middle dose (100,200 mg/kg) has preventive and protective effects in focal cerebral ischemic injury. This protective effect may be related with the increase of Bcl-2 expression and inhibition of Rax expression.
