胰源性14×10 ̄3u磷脂酶A_2(PLA_2)在体外同大鼠中性粒细胞(PMN)培养60min后,细胞对TNF趋化增强,培养10min后上清液中血栓素(TXB_2)含量比正常增高(P<0.01)。前列环素(PGI_2)含量不变。PLA_2激动剂A23187也能显著加强中性粒细胞对TNF的趋化,并伴有TXB_2释放增多(P<0.01)。此外,PLA_2和A23187还显著增强PMN对玻璃珠的粘附活性。使用PLA_2抑制剂二溴苯乙酮(PBPB)和PLA_2多克隆抗体可抑制外源性PLA_2对PMN趋化和粘附的增强作用,但对A23187的调节作用无效。以上结果表明PLA_2激活及其代谢产物可能介导PMN趋化和粘附作用。
Rat PMN were preincubated with purified pancreatic 14×10 ̄3u PLA_2(100ng/ml)and cal-cium ionophore A23187 (10nmol/L)for 60 min to investigate the effects of PLA,on PMNchemotaxis and adherence.The enhancement of chemotaxis towards TNF in agarose and in-crease in release of TXB_2 from PMN were observed,Preincubation of PMN with arachidonicacid(AA),a PLA_2 metabolite,also resulted in TXB_2 release.In addition,PLA_2 and A23187also obviously enhanced PMN adhesion to glass beads(2mm).P-bromophenacyl bromide(PBPB),an inhibitor of PLA_2,at the concentration of 5μmol/L blocked the enhancing effectof PLA_2 and A23187,while 10μmol/L AA could reverse the effect of PBPB.The antiserum ofpancreatic PLA_2 had significant inhibitory effect on PLA_2-primed PMN chemotaxis and adher-ence, but no effect on A23187-primed PMN activation.The results indicate that PLA_2activation may promote or prime the chemotaxis and adherence of PMN by the generation of AAmetabolites.
