肝细胞癌中TRAILR2、AFP、NF-κBp65的表达及其相互关系 被引量:1

摘要 目的探讨肿瘤坏死因子相关凋亡诱导配体(TRAIL)受体TRAILR2、甲胎蛋白AFP和核因子NFκBp65在肝细胞癌(hepatocellularcarcinoma,HCC)凋亡中的作用及相互关系。方法采用免疫组织化学方法检测TRAILR2、AFP、NFκBp65在40例HCC及12例正常肝组织中的表达变化。结果TRAILR2和NFκBp65在40例HCC和12例正常肝组织中均表达,但是HCC组的表达明显增强,两组间表达具有显著性差异(P<0.01);40例HCC中有7例不表达AFP,其余33例可见明显表达,正常肝组织组不表达AFP,两组间表达差异有显著性(P<0.01);HCC中TRAILR2的表达与AFP的表达呈负相关(r=0.464,P<0.01),与NFκBp65的表达也呈负相关(r=0.648,P<0.01)。结论TRAILR2、AFP、NFκBp65与HCC的凋亡有关,AFP抑制BCC凋亡与TRAILR2表达下调有关,AFP下调TRAILR2表达可能是通过NFκBp65的间接作用。 Objective To study the effect and the relationship of TNF-related apoptosis inducing ligand (TRAIL) receptor TRAILR2, alpha-fetoprotein (AFP) and nucleus factor-kappa B (NF-κB) p65 on apoptosis of hepatocellular carcinoma (HCC). Methods By using immunohistochemical technique, the expression of TRAILR2, AFP and NF-κBp65 was detected in 40 HCC and 12 normal liver tissue samples. Results Both TRAILR2 and NF-κBp65 were expressed in 40 HCC and 12 normal liver tissue samples, while their expression was much stronger in the HCC group than in the normal liver tissue group with a significant difference between the two groups (P〈0.01). No expression of AFP was detectable in 7 out of 40 HCC samples, while that was detected obviously in the others. AFP was not expressed in the normal liver tissue samples. There was significant difference between the two groups (P〈0. 017. In HCC group, the expression of TRAILR2 was negatively correlated with both the expression of AFP (r = -0. 464, P〈0. 01 ) and the expression of NF-κBp65 (r = -0. 648, P〈 0. 01). Conclusion All TRAILR2, AFP and NF-κBp65 were relevant to apoptosis of HCC. The effect that AFP suppressed apoptosis of HCC was related to the down-regulation of TRAILR2 expression. What AFP down-regulated TRAILR2 expression may be an indirect effect through NF-κcBp65.
