摘要
本实验用ApaⅠ,BglⅠ,BglⅡ,ClaⅠ,EcoRⅠ,EcoRⅤ,HindⅢ,HpeⅠ,PstⅠ,PvuⅠⅡ,SacⅠ,SalⅠ等12种限制性内切酶分析猪獾和黄鼬的mtDNA限制性片段,并用双酶解法构建限制性内切酶图谱。结合以往积累的资料,我们对哺乳动物mtDNA限制性位点在远缘物种间的保守性和变异性进行了初步讨论。
The mitochondrial DNAs (mtDNAs) from hog-badger (Arctonyx collaris) and siberianweasel (Mustela sibirica) liver were digested with 12 restriction endonucleases, Apa Ⅰ, Bgl Ⅰ,Bgl Ⅱ, Cia Ⅰ, EcoR Ⅰ, EcoR Ⅴ, Hind Ⅲ, HPa Ⅰ, Pst Ⅰ, Pvu ⅠⅡ, Sca Ⅰ, Sal Ⅰ. The mtDNAs were also cleaved by double-enzyme digestions with the above mentioned enzymes.Restriction maps for hog-badger and siberian weasel were made according to the numberand size of those fragments obtained by single and doubledigestions. Combining the previous data, the conservation and variation of mtDNA restriction sites of mammals among far ranged species were also discussed.
