摘要
目的观察肾络通对5/6肾切除肾衰竭大鼠残余肾组织纤溶酶原激活物抑制物-1(PAI-1)蛋白表达的影响。方法5/6肾切除制备肾衰竭大鼠模型,随机分为假手术组、模型组、肾络通组和缬沙坦组,分别灌胃给药16周取材,应用蛋白质印迹法检测肾组织PAI-1蛋白的表达。结果模型组与假手术组相比,PAI-1蛋白的表达明显增高(P<0.01);肾络通组PAI-1蛋白表达较模型组显著降低(P<0.05),与缬沙坦组比较无明显差异。结论肾络通可下调慢性进展性纤维化肾组织PAI-1蛋白表达,改善纤溶系统的失衡,延缓5/6肾切除肾衰大鼠的病程进展。
Objective To observe the influence of Shenluotong on the protein expression of plasminogen activator inhibitor 1 (PAI-1) in kidney remaining tissue in rats with kidney failure induced by 5/6 of nephrectomy, nehtod The rat model of kidney failure was established by 5/6 of nephrectomy and the sham-operation group, model group, Shenluotong group and Valsartan group were set up and treated orally respectively for 16 weeks. The expression of PAI-1 protein in kidney tissue were detected by Western blotting method in different groups. Result The expression of PAI-1 protein was increased significantly in the model group compared with that in the sham -operation group (P 〈0. 01 ), and that in the Shenluotong group was decreased significantly (P 〈 0. 05 ) without significant difference compared with that of the Valsartan group. Conclusion Shenluotong can down-regulate the expression of PAI-1 protein in kidney tissue of chronic developing fibrosis, relieve the unbalance of fibrinolysic system, and delay the development of rat kidney failure after 5/6 of nephrectomy.
