从大鼠活体取背根神经节及交感神经节的方法 被引量:11

摘要 目的探讨取活体的大鼠背根神经节及交感神经节的有效方法。方法Wistar大鼠62只,经戊巴比妥腹腔麻醉,取背根神经节时,大鼠为俯卧位,分离脊柱两旁肌肉,行椎板切开,打开椎间孔,分离并挑出背根神经节;取交感神经节时,大鼠为仰平卧位,剪开右侧胸腔,辨认位于脊柱旁的胸段交感干,分离胸膜后的交感神经节。结果实施背根神经节取出术41例,经该手术方法取得的背根神经节所行细胞培养全部成功;实施交感神经节取出术21例,经该手术方法取得的背根神经节所行细胞培养成功19例,另2例因污染失败。结论采用上述方法,可以成功地获得活体的背根神经节和交感神经节,能够保持神经节细胞良好的活性。 Objective To describe a practical and reliable surgical procedure to dissect dorsal root ganglions and sympathetic ganglions from living rats. Methods Sixty two male Wistar rats were anesthetized by intraperitoneal injection of pentobarbital. When stripping dorsal root ganglions, the rat was in pronation. The paravertebral muscles were detached, and after careful laminectomy the intervertebral foramen was opened, thus the dorsal root ganglion can be exposed and dissected. When stripping sympathetic ganglions, the rat was in supine. After opening the right thoracic cavity, the sympathetic trunk should be found, then to expose and dissect the sympathetic ganglions. Results Operation to dissect dorsal root ganglions was performed on 41 rats. The cell culture of ganglion cells succeeded in all 41 cases. Operation to dissect sympathetic ganglions was carried out on 21 rats. The cell culture of ganglion ceils was successful in 19 cases and failed in 2 cases because of contaimination during cell culture. Conclusion The above described surgical procedures are useful and reliable to obtain dorsal root ganglions and sympathetic ganglions from living rats, with well kept viability and activity of the ganglion ceils.
