摘要
用基质辅助激光解吸附飞行时间质谱(MALDI-TOF-MS)法分析测定了人血清白蛋白。结果表明,全军血液制品研究中心应用Rivanol和Cohn's两种方法分离提取的人血清白蛋白,其分子量与理论计算值相吻合,其纯度与市售标准人血清白蛋白相一致,用MALDI-TOF-MS法分析鉴定人血清白蛋白,准确、简便和灵敏(仅需PM浓度样品),且重复性好,日内和日间差的变异系数均小于0.03%,是SDS-PAGE电泳法所无法比拟的。
The matrix-assisted UV laser desorption ionization time of flight mass sperctra are reported for human albumin which isolated from healthy human blood by Rivanol and Cohn's methods.α-cyano-4-hydroxycinnamic acid is used as matrix in analysis. The RSD within day and between days for the molecular mass determination of human albumin are less than 0. 03 %. The experimental results indicate that MALDI-TOF-MS can be used as a routine method for characterization of human albumin.
