实验性近视眼后极部巩膜胶原和蛋白多糖水平的改变 被引量:5

摘要 目的:研究豚鼠形觉剥夺性近视眼后极部巩膜胶原和蛋白多糖水平的变化,探讨其在哺乳类动物近视眼发生机制中的作用。方法:出生2~3wk的断乳花色豚鼠20只,随机均分成2组。遮盖组右眼予不透明眼罩遮盖2wk,去遮盖组右眼遮盖2wk后去遮1wk。左眼开放为自身对照眼。于实验开始及结束时检影、测眼轴,达规定时限后处死动物,取后极部巩膜,行RT-PCR反应,检测Ⅰ型胶原和decorin核心蛋白mRNA的表达。试验数据采用配对t检验和方差分析进行统计学处理。结果:遮盖组实验眼的后极部巩膜Ⅰ型胶原和decorin核心蛋白mRNA表达水平下降(3.497±0.059→3.057±0.180,1.108±0.147→0.927±0.161,P<0.01),去遮盖组实验眼后极部巩膜Ⅰ型胶原和decorin核心蛋白mRNA表达水平升高(3.657±0.099→3.874±0.058,1.185±0.096→1.253±0.299,P<0.01),组内比较均有统计学意义。结论:豚鼠形觉剥夺性近视眼后极部巩膜Ⅰ型胶原和decorinmRNA表达显著降低,去除遮盖后其表达上调,提示Ⅰ型胶原和decorin共同参与了形觉剥夺性近视眼的发生。 AIM: To investigate the changes of collagen and decorin level in the posterior sdera of Guinea Pig eyes induced by form-deprivation and their association with the pathogenesis of mammal myopia. METHODS: Two groups of Guinea Pigs, with 10 animals each, were included in the study. Animals in the monocular-deprived (MD) group were subjected to 2wk of form deprivation, and those in the MD/recovery group were treated the same way followed by lwk of recovery. Form deprivation by monocular eyeshade was introduced between 2 and 3wk of age in all animals. The contralateral, untreated eyes of the animals served as interocular controls. The refraction and the axial eye length of all animals were measured at the beginning and the end of the treatment period. All animals were sacrificed and eyes enucleated at the end of study. The posterior sdera was dissected from the eye and the expressions of decorin core protein mRNA and collagen Ⅰ mRNA were assayed by RTPCR. The data was analyzed by means of paired t-test and analysis of variance (ANOVA). RESULTS: There were significant differences in the relative level of collagen Ⅰ and decorin core protein mRNA between the MD eyes and interocular controls (P〈0.05) which were also seen between the recovered eyes and interocular controls. CONCLUSION: Collagen Ⅰ and decorin level in the posterior sclera of Guinea Pig eyes decrease significantly upon form deprivation, and can be restored by recovery. It is suggested that they both might be involved in the pathogenesis of myopia.
