米非司酮和利洛司酮对人早孕绒毛、蜕膜分泌功能的影响 被引量:3

Effects of mifepristone and lilopristone on the endocrine function of chorionic and decidual cells during early human pregnancy

摘要 目的比较米非司酮和利洛司酮对人早孕绒毛、蜕膜分泌功能的影响。方法用放射免疫法测定体外培养的人早孕绒毛、蜕膜组织细胞培养液中的人绒毛膜促性腺激素(hCG)、孕酮(P)、雌二醇(E2)、前列腺素F2α(PGF2α)的浓度。结果不同浓度的米非司酮和利洛司酮作用48 h后,绒毛和(或)蜕膜组织培养液中hCG、P、E2的水平均明显低于对照组(P<0.05,P<0.01);PGF2α水平明显增高(P<0.01)。利洛司酮作用后,绒毛和(或)蜕膜组织培养液中hCG、E2和P的水平均低于米非司酮组(P<0.05,P<0.01),PGF2α水平高于米非司酮组(P<0.05,P<0.01).hCG、E2和P的水平与米非司酮和利洛司酮的浓度呈负相关(P<0.01,P<0.05),PGF2α的水平与米非司酮和利洛司酮的浓度无明显相关性。结论利洛司酮和米非司酮均明显抑制离体人早孕绒毛和蜕膜组织细胞的内分泌激素分泌,刺激前列腺素的合成与释放,呈剂量依赖关系;利洛司酮的作用强于米非司酮。 Objective: To evaluate the effects of antiprogestin mifepristone and lilopristone on the endocrine function of chorionic and decidual cells in vitro. Methods. Decidual and chorionic tissue cultures were treated with different concentrations of mifepristone or lilopristone for 48 h, and the levels of hCG, progesterone (P) and estradiol (E2) in the culture medium were determined by radioimmunoassay. Results. Mifepristone or lilopristone at various concentrations used significantly decreased the levels of hCG, P and E2 (P〈0. 01 to P〈0. 05) and increased the levels of PGF2α (P〈0. 01) in the chorionic and decidual tissue cultures. The levels of hCG, P and E2 in the chorionic and decidual tissue cultures treated with lilopristone were lower than those treated with mifepristone (P〈0. 01 to P〈0. 05) while PGF2α levels were higher than those treated with mifepristone (P〈0. 01 to P〈0. 05). There were negative correlations between the levels of hCG, P, E2 and the concentration of mifepristone and lilopristone (P〈0. 01 to P〈0. 05). Conclusions. The results indicate that mifepristone and lilopristone inhibit the endocrine function of chorionic and decidual tissue significantly and stimulate the synthesis of prostaglandin in a dose-dependent manner, and the effect of lilopristone is greater than that of mifepristone.
