清胰汤对胰腺炎肺损伤大鼠Caspase-1及相关细胞因子表达的影响 被引量:1

Influence of Qingyitang(清胰汤) on caspase-1 and cytokines in acute lung injury induced by seere acute pancreatitis in rats

摘要 目的:研究清胰汤对实验性胰腺炎肺损伤半胱氨酸蛋白酶-1(caspase-1)及其相关细胞因子表达的影响。方法:采用15g/L去氧胆酸钠逆行注入胰胆管诱发胰腺炎肺损伤大鼠模型,通过自动生化分析仪检测血清淀粉酶;采用ELISA法测定血清IL-1β水平、RT-PCR检测肺组织Caspase-1 mRNA的表达;免疫组织化学法检测肺组织Caspase-1蛋白的表达。通过检测肺组织髓过氧化物酶(MPO)活性、肺湿/干重比值和肺组织病理切片判定肺损伤程度。结果:胰腺炎肺损伤各组血清淀粉酶和IL-1β水平显著升高,清胰汤及善宁两治疗组血淀粉酶和IL-1β水平明显下降。胰腺炎肺损伤组Caspase-1mRNA和Caspase-1蛋白表达显著上调,清胰汤治疗组和善宁治疗组的表达显著下调。两治疗组肺组织MPO活性减低、肺湿/干重比值下降、肺组织损害程度明显减轻。结论:清胰汤通过下调Caspase-1及其激活的细胞因子IL-1β的表达起到治疗胰腺炎肺损伤作用。 Objective: To study the curative effect of Qingyitang on acute lung injury (ALI) induced by severe acute panereatitis (SAP) in rats. Methods: Forty SD rats were randomized into four groups: sham operated group (group SHAM, n = 10), ALI models (n = 10), ALI + Sandostatin(group SS, n = 10), ALI + Qingyitang(group QYT, n = 10). ALI during SAP was induced by retrograde infusion of 15 g/L sochum deoxycholate into the bili- pancreatic duct in SD rats. All rats were killed 24 hours after operation and treatment. Serum amylase was measured by an automated HITACHI analyzer. Myeloperoxidase(MPO) in pulmonary tissue was measured by enzyme chemistry assay. Serum IL-1β protein was measured by ELISA. Expressions of caspase-1m RNA and protein in pulmonary tissue were detected by RT- PCR and immunohistochemistry. The severity of pulmonary injuries was evaluated by MPO, the ratio of wet/dry lung tissue and pathological change of rats pulmonary tissue. Results: The serum amylase and IL-1β protein levels were increased significantly in group ALI, and those were decreased significantly in group QYT and group SS(P 〈 0.01). The expression of caspase-1mRNA and caspase-1 protein were highly up-regulated in group ALI comparing with group SHAM (P 〈 0.01), and those were down-regulated in group QYT and group SS(P 〈 0.01). The level of MPO, the ratio of wet/dry lung tissue and pathological change of rats pulmonary tissue were increased significantly in group ALI comparing with group SHAM (P 〈 0.01), and those were decreased in group QYT and group SS(P 〈 0.01). Conclusions: Qingyitang can protects ALI induced by SAP with decreasing expression of caspase-1 and release of IL-1 in rats.
