摘要
目的:观察通脉颗粒对嘌呤霉素氨基核苷肾病大鼠纤维蛋白降解产物(FDP)、D-二聚体(DD)、纤溶酶原(P lg)及丙二醛(MDA)的影响。方法:颈静脉插管缓慢注入嘌呤霉素氨基核苷(PAN,50mg/kg)并于此后13、16、19天尾静脉追加注射PAN5mg/kg制备大鼠肾病模型。于造模后第二天开始灌胃给药,连续30天,收集造模后第5、10、15、20、25和30天24小时尿测定尿蛋白排泄。于第31天处死全部大鼠,取血和肾组织进行相关的检查,同时监测FDP、DD、P lg以及MDA等指标。结果:通脉颗粒在减少蛋白尿,改善肾组织病理损害的同时,能够调节FDP、DD、P lg的纤溶活性及清除MDA。结论:通脉颗粒对嘌呤霉素氨基核苷大鼠肾病模型的治疗作用,可能与改善纤溶功能和促进过氧化脂质的清除有关。
Aim: To observe effects of Tongmai Granule on fibin degradation product (FDP), D-Dimer(DD), plasminogen(Plg) and maleic dialdehyde(MDA) levels in puromycin amino nucleoside (PAN)-induced nephropathy rats. Methods: Sixty Wistar rats were randomly divided into six groups (n = 10, in each group): normal group, model group, Monopril group(0.833mg/kg), low-dosage group (treated with Tongmai granule 0.55g/kg), middle-dosage group(treated with Tongmai granule 1.1 g/kg) and high-dosage group (2.2 g/kg). All the rats were injected with PAN (50 mg/kg) via jugular vein except normal rats. After the injection, PAN (5mg/kg) was injected again to the rats via caudal vein on 13, 16 and 19 days. After the nephropathy model was established, the normal group was given with distilled water and the other groups with corresponding drugs by gavage each day for 30 days. Urine was collected from the rats on days 5, 10, 15, 20, 25 and 30 after the model established. The amount of urinary protein was determined. On day 31, all rats were sacrificed for blood biochemistry and light microscopy of the kidneys. The levels of FDA, DD, Plg and MDA in serum were measured. Results: Compared with the model group, Tongmai granule significantly lowered urine protein excretion, increased FDP, DD, Plg fibrinolytic activities (P 〈 0.05) and eliminated MDA (P 〈 0.001). Conclusion: Tongmai granule could ameliorate the pathological changes in the PAN-induced nephropathy rats, which may be associated with improving fibrinolytic function and clearing away lipid peroxides.
