强骨宝对糖尿病性骨质疏松大鼠骨胶原代谢的影响 被引量:5

Experiment on the effect of QIANGGUBAO on metabolism of bone collagen in the diabetic osteoporosis rats

摘要 目的研究糖尿病性骨质疏松(DOP)模型大鼠骨胶原代谢状况及强骨宝方的影响。方法采用四氧嘧啶腹腔注射方法进行大鼠糖尿病(DM)造模,成模后第7周末用DEXA测大鼠骨密度(BMD),取小于正常组平均BMD2.5个标准差的作为DOP模型鼠,再按性别随机分为模型组、阳性对照组及强骨宝组,阳性对照组以消渴丸和阿法迪三(α骨化醇)联合用药,第14周末处死取材,取血液检测IL6、TNFα、IGF1、Ⅰ型前胶原羧基末端前肽(PICP)和Ⅰ型胶原交联羧基末端肽(ICTP)。结果模型组IL6、TNFα、ICTP显著升高,IGF1、PICP显著降低,与正常对照组差异有显著性(P<0.01);经过强骨宝干预后,所有指标均接近正常水平,显著优于阳性对照组。结论DM时骨胶原合成减少及降解加快,易出现OP;强骨宝方可抑制IL6、TNFα的异常表达,刺激IGF1的产生,维持骨胶原代谢的平衡,可以防治DOP,其效果优于消渴丸与阿法迪三联合用药。 Objective To probe into the status of bone collagen metabolism in the diabetic osteoporosis(DOP) rats and the effect of QIANGGUBAO on it. Methods Diabetes mellitus (DM) rats models were established by introperitoneal injection of alloxan. After 7 weeks, the whole body bone mineral density(BMD) was measured by DEXA, and the rats with BMD ≤ 2.5 standard deviation than normal were considered as the DOP rats. According to the gender, the DOP rats were randomly divided into three groups: modeled group, masculine contrast group(treaded with XIAOKEWAN + AIPhaD3) and QIANGGUBAO group (10 rats in each group) all rats were sacrificed at the end of 14 weeks and serum level of IL-6, TNF-α, IGF-1, PICP and ICTP were detected, Results In modeled group, the levels PICP,IGF-1 were lower and IL-6, TNF-α, ICTP higher than the normal contrast group( P 〈 0.01 ). After treatment of QIANGGUBAO, all indexes were similar to the normal group and the effect was superior to masculine contrast group( P 〈 0.01 ). Conculsions Synthesis of bone collagen decreases and its decomposition increases in DM rat which could easily lead to osteoporosis. QIANGGUBAO can maintain the balance of bone collagen metabolism by inhabiting the abnormal expression of IL-6,TNF-α and inducing the production of IGF-1 so as to prevent or treat DOP. And the effect of QIANGGUBAO was superior to the combined treatment of XIAOKEWAN + A1PhaD3.
