摘要
目的:探讨妊娠期糖尿病(GDM)的易感性与HLA-DQA1基因多态性的关系。方法:采用聚合酶链反应-限制性片段长度多态性(PCR-RFLP)技术检测35例GDM患者(GDM组)及35例正常妊娠晚期妇女(对照组)的HLA-DQA1基因多态性频率。结果:GDM组孕妇与对照组孕妇相比较,HLA-DQA1*0201基因频率明显升高(22.86%vs11.43%),差异有显著性(P<0.05)。结论:HLA-DQA1*0201基因在天津及秦皇岛地区汉族GDM患者中频率较高,可作为GDM易感基因的候选基因,未发现GDM的保护基因。
Objective: To investigate the association between polymorphism of HLA-DQA1 and susceptibility of gestational diabetes mellitus (GDM). Methods: This study enrolled 35 cases of GDM and 35 cases of normal pregnant women excluded diseases correlated with HLA gene. Polymerase chain reaction-Restriction fragment length polymorphism (PCR-RFLP) gene typing technique was used to detect HLA-DQA1 alleles. Results: It was observed that gene frequency of HLA-DQAl*0201 increased in GDM group compared with control group (22.86%vs11.43%, P〈0.05,RR=2.84). Conclusion: Gene frequency of HLA-DQAI*0201 significantly increases in GDM at Han ethnic of China. It indicates that alleles HLA- DQAI*0201 may be the susceptible gene of GDM. Which genotype acts as the protective gene is still unknown.
