摘要
目的 建立高效液相色谱(HPLC)法测定血清头孢唑肟浓度。方法色谱柱为μbondaparC18(7.8mm×300mm,5μm),流动相:甲醇-0.01mol·L^-1磷酸二氢钾(20:80),内标为丙酮,检测波长:254nm,柱温:室温,流速:1.0mL·min^-1。结果头孢唑肟的血药浓度在10-20μg·mL^-1范围内与峰面积比有良好的线性关系。血清中头孢唑肟在24h内约降解60.0%。结论 该方法简单,快速,准确,为头孢唑肟的血清浓度监测提供了分析方法。
Objective To set up high performancing liquid chromatography(HPLC) method for determining the serum concentration of ceftizoxime. Methods The chromatographic colu.mn was a μbondapar C18 (7.8 mm × 300 mm, 5μm), the mobile phase consisted of methanol-0.01mol·L^-1 potassium dihydrogen phosphate (20 : 80), the detecting wave-length was 254nm, the flow rate was 1.0 mL·min^-1, and the column was kept at the room temperature. Results The determination limit was 0. 1μg·mL^-1. The linear relationship was fairly good with the amount of ceftizoxime within the range of 10 - 20μg·mL^-1 ( r = 0. 999 5 ). The average rate of recovery was no more than 6.8%. Conclusion The HPLC method is simple, rapid, and accurate for determining the serum concentration of ceftizoxime.
