摘要
以磨盘柿组培苗的无节茎段为外植体,通过3因素3水平正交试验,研究了基础培养基、激素种类及浓度对磨盘柿无节茎段不定芽再生的影响。同时,首次就无节茎段在组培苗上的着生节位、切段大小及接种方式对不定芽再生的影响进行了研究。结果表明:ZT对分化率和平均不定芽数的作用最大,基础培养基次之,IAA的作用最小。IAA对愈伤组织形成的作用大于ZT和基础培养基;组培苗顶端前2节位伸长无节茎段,切成4 mm左右,平放于培养基中可以获得最高的再生效率。组培苗无节茎段在MS(1/2N)+ZT4.0 mg/L+IAA0.1 mg/L培养基中、直接光照培养条件下分化率可达到83.65%,平均不定芽数为2.25。
Using stems without internode taken from "Mopan" persimmon tube seedlings as explants, 3 different kinds of basal culture medium and 2 different kinds of plant hormones with 3 different concentrations were tested with orthogonal design L9 (34)for their optimal effect on adventitious shoot regeneration. At the same time location on the tube seedlings, size and inoculation orientation of stems without internode were studied for the first time. The results showed that the force of ZT concentration was the greatest and the basal culture medium was greater than IAA. But the concentration of IAA was more effective on the forming of callus than basal culture medium and ZT. Using the upper 2 stems without internede of tube seedlings, cutting into about 4 mm in length and laying on the culture medium may enhance the regeneration efficiency of stems without internode. The optimum regeneration medium for stems without internode of "Mopan" persimmon tube seedlings was MS( 1/2N )+ ZT 4.0 mg/L + IAA 0.1 mg/L and cultured in lightness. The rate of regeneration was 83.56 % and the average adventitious shoots number per explant was 2.25.
