摘要
目的观察水溶性氮酮对体外培养的人眼小梁细胞增殖活性和超微结构的影响,寻找氮酮作用于眼部的安全浓度。方法进行人眼小梁细胞的体外培养,MTT法观察不同浓度氮酮作用不同时间后,对小梁细胞增殖的影响;倒置相差显微镜和透射电镜下观察氮酮对小梁细胞形态和超微结构的影响;荧光染色法观察氮酮对小梁细胞活性的影响。结果10^-5~10^-Rg/L的氮酮作用24h不影响小梁细胞的增殖和活性,10^-4g/L的氮酮作用12h不影响小梁细胞的增殖。10^-6-10^-8g/L氮酮作用24h,10^-5g/L氮酮作用12h对人小梁细胞形态和超微结构无影响。荧光染色法显示10^-3g/L和10^-4g/L氮酮作用6h后的细胞呈不同程度变性、坏死。结论10^-6~10^-Rg/L浓度的氮酮不会影响小梁细胞的超微结构、增殖和活性,氮酮有望在眼科广泛应用。
Objective To study the influence of water soluble azone on proliferation and ultrastructure of human trabecular mashwork cells (HTMC) .Methods Human trabecular masbork cells were cultured and passaged in vitro. The cells were treated with different concentrations of azone MTT assay, transmission electron micrope, and acridine orange fluorescent staining were used to observe the influence of azone on the proliferation and ultrastructure of HTMC cells. Results 10 ^- 5 to 10^- 8 g/L of azone treaded for 24 h had no significant influence on the proliferation and vitality of HTMC ceils, 10^-5 g/L of water soluable azone had no significant influence on the proliferation of HTMC cells. 10 ^-6 to 10^ -8 g/L of azone treaded for 12 h had no significant influence on ultrastructure of HTMC cells. Acridine orange fluorecent staining sbowed that HTMC cells treated with 10^-3 g/L or 10^-4g/L of axone for 6h lost its vitality in some degree .Conclusion 10^-6 to 10^-8g/L of water soluable azone had no significant influence on the proliferation, vitality and ultrastructure of HTMC cells. Azone may be. used widely in the eye in the future.
