摘要
目的初步研究血红素氧合酶-1(HO-1)蛋白在股骨头缺血后的表达变化及意义。方法采用中断家兔股骨头血供致股骨头缺血,应用HE染色,免疫组织化学技术来观测正常兔、股骨头缺血后1.5、3、6、12、24 h兔股骨头内HO-1的表达及含量变化。结果正常兔股骨头内HO-1的表达较少;缺血后3、6 h,HO-1在股骨头内的表达增加,主要在成骨细胞和骨细胞中,骨髓细胞中也有表达,缺血后3、6 h阳性细胞的平均灰度值和光密度分别为(80.09±9.92)和(1.16±0.11)、(56.74±11.38)和(1.54±0.17);缺血后12 h HO-1表达强度开始下降,但是下降不明显,阳性细胞平均灰度值和光密度值分别为(93.50±18.36)和(1.21±0.19);缺血24 h表达减弱至近正常对照组。结论兔严重股骨头缺血后引起HO-1表达增加,说明HO-1参与股骨头缺血后股骨头的坏死过程,其可能是机体对股骨头缺血的一种自身恢复机制。
Objective To observe the expression of heme oxygenase-1 ( HO-1 ) after femoral head ischeamia and its significance. Methods Thirty male rabbits were randomly divided into normal group ( n =5) and experimental group ( n =25). The rabbits were given no operation in the normal group and were given ischeamic injury in the experimental group. The femoral head ischeamia animal models were made and rabbits in the experimental group were sacrificed at l. Sh ( n =5), 3h ( n =5) , 6h ( n =5), 12h ( n =5) and 24h ( n = 5 ) after operation. The distribution and content changes of HO-1 in femoral head were observed by means of immunohistochemistry analysis. Results The expression of HO-1 was very low in normal femoral head and 1.5 h experimental group. The expression of HO-1 was increased in femoral head at 3 and 6 hours after iscbeamia and the expression was primarily existed in osteoblasts and osteocytes. The expression mean gray level (GL) and the average optical density (OD) of the positive cells at 3 hours after iscbeamia were ( 80. 09 ± 9. 92 ) and ( 1.16 ±0. 11 ), respectively. At 6 hours after the operation , the HO-1 protein expressed with expression mean GL of(56. 74 ±11.38 ) and OD of ( 1.54 ±0. 17). At 12 hours after the operation , the expression mean GL and OD were decreased to (93.50 ±18.36)and ( 1.21 ±0. 19) respectively. The expression was decreased at 24 hours after operation significantly. There was significant difference between the experimental groups and normal group at 3, 6, 12h after iscbeamia ( P 〈0. 01 ). Conclusion The expression of HO-1 is increased after severe ischaemia in femoral head, which shows that HO-1 may be involved in the necrosis of femoral head after ischaemia and the recovery from ischaemia injury.
