摘要
目的:探讨用多柔比星腹腔注射建立扩张型心肌病模型的可行性。方法:取10周龄雄性SD大鼠(350±50)g,共40只。随机分为4组:对照组、低剂量组、中剂量组、高剂量组,给药组腹腔注射量分别为8、16、24 mg/kg,并于给药前及给药后第2、4、6、8周用心脏超声观察收缩期、舒张末期内径及射血分数;第9周取心肌观察病理形态特征结果:心脏超声检查显示给药8周后中、高剂量组大鼠左心室收缩期末口径及舒张期末口径增大,左心室射血分数下降(P<0.05);组织病理检查符合扩张型心肌典型病理特征,但高剂量组大鼠生存率较低。结论:予以大鼠多柔比星腹腔注射每周2 mg/kg,8周可建立扩张型心肌病实验模型。
Objective: To investigate the feasibility to establish experimental model of dilated cardiomyopathy. Methods: Forty male 10 weeks old SD rats (350 :t: 50) g were randomly divided to 4 groups : control group, low dose group, middle dose group and high dose group. By the toxicity of adriamyein, different doses of adriamyein were given to rats by intraperitoneally injected. The left ventrieular dimensions at endsystole and end-diastole (LVDs, LVDd) and the ejection fraction (EF) were measured by eehoeardio- graphy at the 2^eh, 4^th, 6^th, 8^th weeks. Histopathological characteristics were observed at the 9^th week. Results: LYDD and LVDS in middle dose group and high dose group were significantly increased, and EF were significantly reduced ( P 〈 0.05 ) in these two groups compared with those rats in control group. Cardiac histopat.hological characteristics were similar to those of human dilated cardiomyopathy at the 9^th weeks in middle and high dose groups. Conclusion: SD rats with adriamycin intraperitoneally injected (2 mg/kg,8 weeks may be a desirable experimental model of dilated cardiomyopathy.
