摘要
对健康小鼠肝细胞、实验性肝癌小鼠肝细胞及肝癌实体瘤细胞中琥珀酸脱氢酶、乳酸脱氢酶和细胞色素氧化酶化活力进行测定。发现:1)与健康鼠肝细胞比较,肝癌细胞中琥珀酸脱氢酶比活力显著降低(P<0.01),乳酸脱氢酶、细胞色素氧化酶比活力显著升高(P<0.01)。2)与正常肝细胞比较,荷瘤小鼠肝细胞中乳酸脱氢酶、细胞色素氧化酶比活力显著升高(P<0.01)。琥珀酸脱氢酶比活力无明显变化(P>0.05)。结果提示引起恶性肿瘤细胞强烈糖酵解作用的原因之一可能是其三羧酸循环受到抑制,而氧化磷酸化作用的增强为肿瘤细胞提供了比正常细胞更多的能量。实验结果为用乳酸脱氢酶、细胞色素氧化酶作为恶性肿瘤临床诊断的生化指标提供了依据。
The specific activities of succinate dehydrogenase, lactate dehydrogenase and cytochrome oxydase were identified in three kinds of liver cells of mice: normal liver, liver of the cancer bearing mice and the cancer self. The results show that: 1) The specific activity of succinate dehydrogenase on cancer cells significantly lower than that of the normal (P< 0. 01); the specific activity of lactate dehydrogenase and cytochrome oxydase rose up significantly (P < 0. 01) . 2) Compared with normal liver, the specific activity of lactate dehydrogenase and cytochrome oxydase on cancer bearing mice liver significantly rose up (P < 0 . 01), but, the succinate dehydrogenase had no significant change (P >0 . 05). The results indicated that one of the reasons of the strong glyucolysis in cancer cells may be because of the inhibition of TCA cycle, the high activity of oxydative phosphorylation seems to supply more energy to cancer cells. The results offer a support to establish a biochemical inspection for cancer disease by using the activity identification of lactate dehydrogenase and cytochrome oxydase.
