老年人肺癌组织中鸟氨酸脱羧酶基因表达及其临床意义 被引量:2

摘要 目的:从mRNA和蛋白质水平探讨ODC基因表达和老年人肺癌的关系,为肺癌基因诊断和基因治疗的研究提供理论依据。方法:分别应用RT-PCR和免疫组化方法检测42例老年人肺癌组织和癌旁正常肺组织中ODC mRNA和ODC蛋白的表达,应用Western Blot检测肺癌组织中ODC表达,同时用MTT法观察不同转染组肿瘤细胞的增殖状况,探讨上述指标与肺癌临床病理学特性的关系。结果:RT-PCR显示,老年人肺癌组织中ODC mRNA表达明显高于癌旁正常肺组织(P<0.05),不同类型和分化程度肺癌组织中ODC mRNA水平差异无统计学意义(P>0.05),Ⅲ期肺癌组织中ODC mRNA表达明显高于Ⅰ期+Ⅱ期(P<0.05);免疫组化显示,肺癌组织中ODC蛋白表达明显高于癌旁正常肺组织(P<0.05);不同病理类型和分化程度肺癌组织中ODC蛋白表达未见明显差异(P>0.05),Ⅲ期肺癌组织中ODC蛋白表达较Ⅰ期+Ⅱ期明显升高(P<0.05);Western Blot显示,肺癌组织中ODC蛋白表达明显高于癌旁正常肺组织;MTT法观察生长曲线检测数据表明,ODC的反义RNA表达载体转染组细胞的增殖明显比对照组细胞和空载体转染组细胞缓慢。结论:ODC表达增强与老年人肺癌发生、发展、浸润及转移有关,可作为判断肺癌生物学行为的参考指标;ODC的反义RNA表达载体对A-549肺癌细胞株的生长具有一定的抑制作用。 Objective: To study the relationship between the gene expressions of omithine decarboxylase and lung cancer in the eider from the level of mRNA and protein, and therefore to establish the theoretical basis for gene diagnosis and gene therapy in the future. Methods: RT-PCR technique was used to observe the density of ODC mRNA, and immunohistochemical method to observe the difference of ODC mRNA. Westem Blot technique was used to observe the density of ODC. At the same time, MTT technique was used to observe the multiplication of differenttransfected cells. Results: The expressions of ODC mRNA and protein in lesion of lung cancer were significantly higher than those in contiguous normal tissues ( P 〈 0.05), and they were quitely higher in stage Ⅲ than stage Ⅰand stage Ⅱ ( P 〈 0.05). The growth curve observed by MTT showed that multiplication was slower in transfected cells than in contrasts. Conclusion: ODC gene plays an important role in lung cancer and the up-regulated expression of ODC is related to the occurrence and development of lung cancer. ODC recombinant vectors could inhibit the growth of A-549 lung cancer cell lines.
