摘要
目的:建立测定诺坤复片中雌二醇含量的胶束电动毛细管色谱法(MECC)。方法:采用胶束电动毛细管色谱法,测定条件为:电泳缓冲液为20mmol·L^(-1)硼砂缓冲溶液(pH=9.2),内含40mmol·L^(-1)十二烷基硫酸钠(SDS)及5%甲醇,分离电压为18kV,温度为25℃,空心熔融未涂层石英毛细管柱75μm(id)×35cm(有效长度),检测波长为280nm,进样方式为压力进样,进样时间5.0s,外标法定量。结果:雌二醇的线性范围为25~400μg·mL^(-1)(r=0.9998),平均回收率为100.1%,RSD为1.63%(n=3)。结论:该方法简便、准确、灵敏、适用于雌二醇的质量控制。
Objective:A useful method for determination of estradiol in estrofem tablets by micellar electrokinetic capillary chromatography(MECC) has been developed. Methods:MECC ,the buffer solution was 20 mmol ·L^-1 borate buffer (pH =9. 2) ,containing 40 mmol ·L^-1 sodium dodecyl sulfate (SDS) and 5% methanol. The effective length of 75 μm (id) fused silica capillary was 35 cm and the detective wavelength was 280 nm. The voltage and temperature was 18 kV and 25 ℃ respectively. Sample were injected into the capillary by applying pressure for 5.0 s. The concentrations were quantified by external standard method. Results: The calibration curve of estradiol was linear over the range of 25 -400μg · mL^-1 (r =0. 9998). The average recovery was 100. 1% (n =3). Conclusion:The method is simple, accurate and sensible, and can be used for quality control of the estradiol.
