摘要
目的制备紫杉醇聚乳酸纳米粒。方法采用乳化溶剂挥发法制备紫杉醇聚乳酸纳米粒,以载药量为主要评价指标,选择氢氧化钠用量、超声强度、卵磷脂用量、二氯甲烷体积为考察因素,采用均匀设计试验优化制备工艺。结果4个因素中,卵磷脂用量对载药量影响最大,其次是氢氧化钠用量和超声强度,而二氯甲烷体积的影响很小。优化工艺制备的紫杉醇聚乳酸纳米粒粒径范围为(32.9±12.3)nm,载药量为8.70%,包封率为99.44%。结论该制备工艺简便稳定,具有应用前景。
Objective To prepare the paclitaxel-loaded poly(D, L-lactic acid) nanoparticles (TAX-PLA- NP). Methods An emulsion solvent evaporation technique was developed. Four factors, including the amount of NaOH, sonic intensity, the amount of lecithin and the volume of dichlormethane were selected to establish the optimal conditions by a homogeneous design. The drug loading was introduced as an index for the nanoparticles formulation. Results The four factors affected drug loading differently, in descending order: the amount of lecithin, the amount of NaOH and sonic intensity, the volume of dichlormethane. The average diameter of TAX-PLA-NP was (32.9±12.3 ) nm, and the drug loading and the envelope ratio of TAX-PLA-NP were 8.70% and 99.44%. Conclusion The preparation procedure is good, simple and reproducible.
