摘要
通过单因素实验和正交实验,利用愈创木酚比色法研究了萌发玉米中过氧化物酶的提取pH值、影响过氧化物酶活力的各种因素和过氧化物酶活力测定的最佳条件。实验结果表明,过氧化物酶的最佳提取pH值是6.7;各因素对过氧化物酶活性的影响顺序为萌发时间>温度>pH值>反应时间。萌发玉米种过氧化物酶提取的最佳条件为,温度40℃,pH5.4,萌发时间3d,反应时间5min。
Extraction pH of peroxidase in germinated corn, factors 'affect- ing peroxidase activity and the optimal determination of peroxidase were studied by colorimatric method with single factor and orthogonal experiments. The results showed that the optimal Extraction pH of peroxidase was 6.7, the order of the factors affecting peroxidase activity was : germinating time 〉 reaction temperature 〉 reaction pH 〉 reaction time,the optimal condition for the determination of perooxidase activity was, 40 ℃, reaction pH:5.4, germinating time: 3 days, reacting time: 5 min.
