摘要
目的 建立大鼠血浆中淫羊藿苷的检测方法。方法 采用RP-HPLC法测定大鼠血浆中的药物浓度,色谱柱为Lichrospher C18(4.6ram×250mm,5μm),流动相为甲醇-水-冰醋酸(45:55:0.5v/v),检测波长为270nm,流速为1ml·min^-1。结果 在1~96mg·L^-1范围内淫羊藿苷峰面积与浓度线性关系良好(r=0.9993),最低检测浓度为0.03mg·L^-1,绝对回收率为85.3%~96.6%,准确度为94.3%~101.2%,日内、日间RSD均小于10%。结论 本测定方法灵敏、准确、简便,适合于淫羊藿苷的药代动力学研究。
Objective To develop a simple, sensitive and rapid method for the determination of Icariin in rat plasma. Methods The plasma concentration of Icariin was determined by RP - HPLC method, the analysis involved a 250 mm × 4.6mm column packed with C18 ( 5 μm ) ,the mobile phase consisted of methyl alcohol -water- glacialacetic acid (45:55:0.5 ) ,the detection was performed at UV 270nm. Results A good linearity was obtained in the range of 1 ~ 96 mg/L ( r = 0. 9993 ) , the recovery was between 85.3 ~ 96.6% , the accuracy was between 94.3% ~ 101.2%. The relative standard deviations of within - day and between day were less than 10%, the minimal detectable concentration in rat plasma was 0.03 mg/L. Conclusion The result demonstrated that the method was sensitive, accurate and simple for the determination of Icariin in rat plasma, it was suitable for the pharmacokinetics study of Icariin.
