摘要
目的观察大鼠实验性脑挫伤后CNS内β-APP蛋白表达时空性规律。方法采用自由落体撞击法致大鼠脑挫伤模型,用免疫组化技术观察伤后β-APP蛋白在不同时间(0.5h、2h、6h、12h、24h、3d、7d、14d)的表达情况,并用计算机图像分析技术作定量统计分析。结果(1)皮质内β-APP蛋白在伤后表达呈一定波动性,0.5h表达明显增加后,逐渐下降,12h降至最低,低于对照组,随后再次表达增加,3d达高峰;(2)伤后24h海马区β-APP表达开始逐渐下降,3d降至最低,14d回到对照组水平;(3)齿状回(DG)内β-APP蛋白在伤后12h低于对照组。结论实验性脑挫伤后皮质β-APP表达时序性规律可为脑损伤时间推断提供一个新的参考指标,但海马区及DGβ-APP变化规律在脑损伤时间推断方面价值不大。
Objective To Study the change of β-APP expression in different wound age and site in the experimental traumatic brain injury in rats. Methods The animal model of cerebral contusion was established by impact to the parietal lobe with a free fall weight,β-APP expression in different wound age was observed with Immunohistochemistry. The results were measured quantitatively with computer imaging analysis system. Male Spragure-Dawley rats were divided into 3 groups: normal control, sham operation control and injury group. Results (1)The number of β-APP-positive neurons definitely increase in 0.5 h after brain injury, decline gradually and reach valley bottom in 12h, increase again and reach the peak in 3 d. (2)In hippocampus the number of β- APP-positive neurons definitely increases after 24 h after brain injury, then decline gradually and reach valley bottom in 3d, return to the level of control at 14 d. (3)In dentate gyms (DG) the number of β-APP-positive neurons at 12 h is lower than the control after brain injury.The intensity of the immunoreactivity of β-APP protein changes with time after traumatic brain injury, which might be used for determing injury time, however, the change in hippocampus and DG has little value in determing injury time.
