摘要
目的利用血管紧张素Ⅱ(AngⅡ)刺激心肌细胞造成肥大,观察心肌细胞肥大过程中心肌营养素-1(cardiotrophin-1,CT-1)的表达情况。方法培养原代心肌细胞,给予AngⅡ刺激心肌细胞造成肥大,在不同时间取细胞进行反转录聚合酶链反应(RT-PCR)观察CT-1 mRNA表达,免疫组化法检测CT-1蛋白表达,相差显微镜下观察细胞形态变化。结果经AngⅡ刺激后在相差显微镜下可见心肌细胞面积变大。CT-1 mRNA水平随着AngⅡ刺激时间延长而有增高的趋势,CT-1蛋白表达亦增加。结论AngⅡ致心肌细胞肥大过程中伴有CT-1表达增加,CT-1有可能参与心肌细胞的肥大机制。
Objective To observe the expression of cardiotrophin-1 (CT-1) in hypertrophic cardiomyocytes induced by angiotensin lI (AngⅡ). Methods The cultured cardiomyocytes were treated with Ang lI to induce hypertrophy. In different periods, the mRNA levels of CT-1 were examined by real-time PCR method. The protein expressions of CT-1 were examined by immunohistochemistry method. The morphologic changes of cardiomyocytes were observed under the contrast phase microscope. Results The myocytes were enlarged under the contrast phase microscope. After stimulated by Ang, the mRNA level and protein expression of CT-1 increased in the hypertrophic cardiomyocytes. Conclusions The expression of CT-1 increased in hypertrophic cardiomyocytes stimulated by AngⅡ CT-1 may play a role in cardiocyte hypertrophy.
