摘要
目的阐明维生素C(VC)对低硒(Se)高镉(Cd)饲料饲养大鼠心肌细胞DNA的影响。方法按喂养大鼠的饲料不同分组,用单细胞凝胶电泳技术分别观察普通饲料(对照1),实验构成饲料(对照2),单纯高Cd、高Cd高VC、低Se高Cd、低Se高Cd伴高VC、高Se高Cd、高Se高Cd伴高VC饲料饲养14周后大鼠心肌细胞DNA的改变。结果高Se高Cd伴高VC组大鼠心肌细胞DNA损伤率与对照组比较差异无统计学意义(P>0.05);高Cd高VC组、高Se高Cd组心肌细胞DNA轻度损伤;低Se高Cd高VC组、单纯高Cd组心肌细胞DNA损伤较重;低Se并高Cd组心肌细胞DNA损伤最为严重,损伤率达26.18%(P<0.01)。结论低Se并高Cd可在一定程度上改变心肌细胞的DNA生物学特性,VC可以对抗低Se并高Cd对心肌细胞所致的这种变化。
Objective To elucidate the etiology of DNA impairment of myocardial cells of the rats fed with low selenium (Se) and high cadmium (Cd) fodder. Methods With single cell gel electrophoresis technique, the joint action of Se, Cd and vitamin C (VC) on DNA damage was observed in myocardial cells of the eight groups of rats fed for 14 weeks with regular fodder (control group 1), fodder used in this research but without additive (control group 2), fodder with high Cd, high Cd + high VC, low Se + high Cd,low Se + high Cd + high VC, high Se + high Cd and high Se + high Cd + high VC, respectively. Results Compared with the control, there was no DNA changes observed in high Se + high Cd + high VC group(P 〉 0.05). However,in high Cd + high VC group and high Se + high Cd group, DNA damage of the myocardial cells could be detected clearly; in low Se + high Cd + high VC group and high Cd group, the degree of the DNA damage was more serious than that in the two groups mentioned above;in low Se + high Cd group,DNA was damaged the most seriously and obviously in all the groups studied (damaging rate of the cells reaching 26.18% ). Conclusions It is suggested that Se deficiency and accompanied by overdose of Cd may damage DNA of the myocardial cells of the rats significantly, however, VC may protect myocardial cells against the injury effectively.
