摘要
本实验对根霉RhizopusRA-1淀粉酶进行了分离纯化,经过(NH4)2SO4分步沉淀,DEAE-纤维素离子交换层析和Sephadex G-100凝胶层析等3部纯化,得到了电泳纯的糖化酶和两条紧密相连的液化型淀粉酶带,用SDS-PAGE法测得三种酶都只含有一种亚基,相对分子质量分别为78000,48000,50000。
Amylases of Rhizopus RA-lare separated and purified by (NH4)2SO4 fractional precipitafion,DEAE-cellulose ion-exchange chromatography and gel G-100 filtration.It was determined by PAGE that the amylase shows glucoamylase line and two α -amylase lines, detemined by SDS-PAGE the three molecular weights are about 78kD,48kD and 50kD.
