摘要 目的研究体外缺氧对乳腺癌细胞系MCF7浸润能力及其细胞表面金属蛋白酶MMP-2、丝氨酸蛋白酶u-PA受体u-PAR和细胞基质蛋白FN的表达的影响。方法本实验模拟体外缺氧环境,观察缺氧对乳腺癌细胞MCF7浸润穿透Metrigel的能力;以及采用半定量RT-PCR检测细胞表面MMP-2、u-PAR和FN表达情况。结果缺氧状态下MCF7细胞的浸润能力明显增强;其表面的MMP-2、u-PAR、FN基因表达升高。结论缺氧可以上调与乳腺癌浸润转移有关的基因的表达,缺氧条件促进乳腺癌细胞的浸润转移。 Objective To study the effect of hypoxia on breast cancer cell MCF7 invasion through Matrigel and the expression of MMP-2, u-PAR, FN mRNA level in vitro. Methods In vitro Matrigel invasion of MCF7 cells and the expression levels of MMP-2, u-PAR and FN in the MCF7 cells in hypoxic condition were analyzed. Results Hypoxia promoted MCF7 cells invade through Matrigel. MMP-2, u-PAR and FN mRNA levels were up regulated under hypoxic conditions. Conclusion Hypoxia promotes breast cancer cell MCF7 invasion by inducing the up-regulation of MMP-2, u-PAR and FN expression.
