摘要
目的观察小鼠出生后肾脏皮质发育过程中增殖细胞核抗原(proliferating cell nuclear anti-gen,PCNA)的表达及细胞凋亡的特征,探讨出生后小鼠肾脏皮质发育过程中细胞增殖与凋亡的规律及其关系。方法应用免疫组织化学技术和原位末端标记法(TUNEL法)分别检测出生后1、3、7、14、21、28和70天小鼠肾脏皮质中PCNA阳性的细胞和凋亡细胞。结果(1)小鼠生后肾脏皮质发育中PCNA的表达有一定的规律,早期PCNA阳性表达丰富,随着肾脏发育成熟表达逐渐减弱直至成年基本消失。(2)小鼠出生后肾脏皮质发育中也存在着大量细胞凋亡现象,且凋亡高峰一般出现在增殖高峰之后。结论细胞增殖与凋亡在小鼠生后肾脏皮质发育的整个过程中普遍存在,早期细胞增殖旺盛,增殖高峰之后出现凋亡高峰,晚期两者活动均减弱。
Objective To observe the expressive characteristics of proliferating cell nuclear antigen ( PCNA ) and apoptosis, and investigate the activity of cell proliferation and apoptosis in the mouse kidney cortex during postnatal development. Method The expression of PCNA and apoptosis were examined in the mouse kidney cortex after birth 1,3, 7, 14, 21,28 and 70 days by immunohistochemical technique and Tclt-mediated clUTP nick end labelling ( TUNEL). Results ( 1 ) After birth, the activity of cell proliferation was prominent. At the early stage, the PCNA-p6sitive cells were abundant. Proliferative activity tended to decrease gradually as the renal developmental stage proceeded and eventually varnished. (2) Apoptotic cells were present too. And cell death peaked later than cell proliferation. Conclusion Cell proliferation and apoptosis exist universally in the mouse kidney cortex during postnatal development. In the early stage, the positive rate of PCNA was high, and the positive rate of apoptosis was high later, and then the activities of them were both weak.
