摘要
目的:观察脊髓慢性受压不同时间以及手术减压后不同时间受压节段神经细胞凋亡的变化。方法:将56只雄性山羊随机分为对照组(8只)、颈脊髓受压3个月组(24只)和6个月组(24只),同时将受压迫3、6个月组再分为减压术前及术后1、3个月组(每组8只)。实验动物通过颈椎前路钛合金螺钉和后路钢板置入产生慢性压迫。各组在相应时间点处死动物后取压迫节段脊髓标本,应用TUNEL技术和免疫组织化学方法检测受压脊髓不同部位神经细胞的凋亡情况。结果:脊髓慢性受压3、6个月后在受压节段不同区域均可见凋亡细胞,受压6个月组较受压3个月组有更多的凋亡细胞数(P<0.05)。后角胶质细胞凋亡变化幅度最大,而后角神经元凋亡变化幅度最小。减压手术后1个月脊髓各区凋亡细胞均减少,减压后3个月时减少更为明显(P<0.05)。结论:颈脊髓慢性受压后不同时间在脊髓不同区域出现不同数量的凋亡细胞,压迫时间越长凋亡细胞数越多;减压手术后脊髓受压区域凋亡细胞数目开始减少,术后时间越长凋亡细胞数目越少。
Objective:To observe the effect of chronic cervical spinal cord compression and decompression on the neuron apoptosis at different time point.Method:Fifty six goats with 20-24 months old (weights :13- 17kg,mean:15kg) were randomly divided into five groups:control group(n=8),3 months compression(n=24) and 6 months compression (n=24) group,every compressional group were subdivided into 1 month and 3 months group after decompression respectively.All experimental animals underwent cervical spinal cord compression resulted from anterior Titanium screw and posterior plate implanted.Decompression was performed at 3 or 6 months after compression.The number of cells labeled by TUNEL (Terminal deoxynucleotidyl-transferase-mediated deoxyuridine triophosphate-biotin nick end labelin) staining were counted under Olympus BX51 microscorp (400x),cell counting at the anterior,lateral,and posterior columns as well as anterior and posterior horns in the compressed region was performed. Result:Apoptosis was evidenced in each group at 3 and 6 months after chronical cervical spinal cord compression,the apopcell number of at 6 months after compression was bigger than that at 3 months(P〈0.05). In posterior horns,the apopcell number showed more changes than in other groups.The apopcell number decreased in each group after 1 and 3 month of decompression,with more remarkable at 3 month after decompression(P〈0.05). Conclusion:There are significant in terms of apopcell number between 3 month groups and 6 month groups after chronical cervical spinal cord compression,the apopcell number are increasing with the time which is conversed after decompression.
