孕妇尿液表皮生长因子的分离、纯化及活性检测

摘要 孕妇尿液蛋白丙团粉粗提液经Bio-gelP-10凝胶层析获得五个组分,经免疫点印渍分析后,证明其中组分Ⅲ的活性最高,该组分再经SephadexG—15凝胶层析,又得到A、B、C和D四个组分,hEGF活性主要分布在组分A中。HPLC的分析结果表明组分A的分子量在6,000左右。生物活性研究证明,样品可以促进初生幼鼠的睁眼和萌牙。本文建立了免疫点印渍方法并应用于hEGF的活性检测。 The study presented was about the preparation of hEGF from pregnant's urine. The isolation procedure involved two-step chromatography. In the first step, the main active fractionwas fraction ill of the five fractions from Bio-gel P-10 column chromatography, the active fraction went again on sephadex G-15 and four fractions was obtained, but the activity concentrated was on peak A. The molecular weight of hEGF sample was about 6, 000 identified byHPLC. Immuno-Dot blotting was applied to detect of hEGF activity and met with success. Themethod was simply, fast and sensitive. The hEGF sample had obvious activity to stimulate eyelids open and tooth eruption of new born mice.
