活血化瘀汤对大鼠骨折愈合中转化生长因子β1 mRNA与蛋白表达及软骨内成骨的作用 被引量:22

摘要 目的:观察活血化瘀汤对实验性大鼠骨折愈合过程中转化生长因子β1表达的影响。方法:实验于2005-04/12在福建中医学院骨伤研究所实验室完成。选择健康3个月龄雄性SD大鼠60只,建立闭合骨折髓内固定动物模型后随机数字表法分为活血化瘀汤组(n=30)、生理盐水组(n=30)。每组又分为骨折后3,5,7,14,21d5个时相点,每个时相点6只。在大鼠麻醉苏醒后开始灌服活血化瘀汤(由蒲黄9g,姜黄4.5g,当归9g,泽兰6g,茜草9g,三七6g,红花1.5g,莪术6g,生地9g,赤芍9g,紫苏9g,甘草3g等组成,由福建中医学院屏山制药厂协作制备)15mL/(kg·d)(按l0倍成人剂量换算),分两次,生理盐水组在相同条件下灌服等体积生理盐水,分别于骨折后3,5,7,14,21d取骨折区组织,通过反转录-聚合酶链反应,酶联免疫吸附法进行分析转化生长因子β1的表达。结果:纳入大鼠60只,均进入结果分析。①转化生长因子β1的表达在骨折后第3天有一个初期峰值;在第7天会出现一个低谷;而在骨折后第2周,转化生长因子β1的表达会再次升高,达到第2个峰值,也是最高表达期;其后的表达逐渐降低。②骨折后3,5,7,14,21d活血化瘀汤组转化生长因子β1mRNA的表达高于生理盐水组(分别为0.437±0.073,0.292±0.103;0.368±0.099,0.245±0.052;0.360±0.082,0.281±0.076;0.783±0.289,0.461±0.163;0.526±0.120,0.453±0.085,P<0.01)。③骨折后3,5,7,14,21d活血化瘀汤组转化生长因子β1蛋白的表达高于生理盐水组(分别为36.033±2.244,32.879±3.423;34.442±2.197,32.691±1.243;33.383±1.688,32.073±1.109;40.918±4.102,38.287±2.694;34.688±3.746,33.381±0.499,P<0.01)。结论:活血化瘀汤使转化生长因子β1的表达增强,促进了骨折愈合。 AIM: To observe the effects of Huoxue Huayu Tang on mRNA and protein expression of transforming growth factor beta 1 (TGF-β1) in the fracture healing of experimental rats. METHODS: The experiment was conducted in the laboratory of Institute of Orthopaedics and Traumatology in Fujian College of Traditional Chinese Medicine from April to December in 2005. Kirschner wires were fixed in the bone marrow of the midshaft tibia with standardized closed fracture in 60 healthy male SD rats of 3 months old to establish the fracture models. The rats were randomly and equally divided into treatment group and saline group. And each group was assigned into 5 time points (3, 5, 7, 14 and 2! days after fracture) containing 6 rats on each point. The rats after anesthesia of treatment group were administrated with Huoxue Huayu Tang (containing 9 g cattail pollen, 4.5 g turmeric, 9 g angelica, 6 g hirsute shing bugleweed herb, 9 g india madder root, 6 g sanchi, 1.5 g safflower, 6 g zedoary, 9 g rehmannia dride rhizome, 9 g red peony root, 9 g perilla and 3 g glycyrrhizin, assistantly prepared by Pingshan Pharmaceutical Factory of Fujian College of Traditional Chinese Medicine), 15 mL/kg per day in two administrations, while the rats of saline group were treated with equal volume of saline. The specimens of bony calluses were collected at 3, 5, 7, 14 and 21 days after fracture to detect the expression of TGF-β1 by reverse transcription-polymerase chain reaction and enzyme-linked immunosorbent assay. RESULTS: All the 60 rats were involved into the result analysis. ①The expression of TGF-β1 increased to the first peak on the 3^th day after fracture, but appeared a low point on the 7^th day, and returned to peak at the second week, then gradually decreased later.②The expression of TGF-β1 mRNA was higher in treatment group than in saline group (0.437±0.073, 0.292±0,103; 0,368±0,099, 0,245±0,052; 0,360±0.082, 0,281±0.076; 0.783±0,289, 0.461±0,163; 0,526±0.120, 0.453±0.085, P 〈 0,01),③The expression of TGF-β1 protein was higher in treatment group than in saline group (36.033±2,244, 32.879±3,423; 34,442±2,197, 32,691 ±1,243; 33,383±1.688, 32.073±1,109; 40,918±4,102, 38,287±2.694; 34,688±3,746, 33,381±0,499, P 〈 0,01), CONCLUSION: Huoxue Huayu Tang may increase the expression of TGF-β1 in bony callus of rats with early fracture, and thus promote the fracture healing,
