辛伐他汀对糖尿病大鼠肾小球NF-κB ICAM-1表达的影响 被引量:1

摘要 目的研究观察核因子κB(NFκB)、细胞间黏附分子1(ICAM1)在糖尿病大鼠肾小球的表达及辛伐他汀干预治疗的情况,初步探讨大鼠糖尿病肾病的发病机制。方法3组大鼠:正常对照(NC)组、糖尿病(DM)组、辛伐他汀治疗(D)组,分别于实验第4、8周末,检测血糖、血脂、肌酐清除率(Ccr)、24h尿微量白蛋白等;免疫组织化学染色检测肾小球NFκB、ICAM1的表达。结果4周时,DM组及D组Ccr、24h尿微量白蛋白、NFκB、ICAM1表达均显著高于NC组,D组与DM组上述指标间差异无统计学意义;8周时,DM组及D组Ccr、24h尿微量白蛋白、NFκB、ICAM1表达均显著高于NC组,D组上述指标明显低于DM组。结论辛伐他汀可引起NFκB、ICAM1表达下调,而发挥抗炎、肾脏保护作用。 Objective To investigate the effects of simvastatin on nuclear factor (NF)-κB and intercellular adhesion molecule (ICAM)- 1 expression on glomeruli of diabetic rats,and explore the possible mechanisms. Methads Experimental rats divided into three groups:NC,DM and D. After weeks 4 and 8,Ccr,serum cholesterol and UAE were measured. Expressions of NF-κB and ICAM-1 were detected by immunohistochcnnical staining. Results On week 4, Ccr, UAE and expressions of ICAM-1 and NF-κB in D group were not significantly lower than those in DM gourp on week 8. Ccr,UAE,and expressions of NF-κB and ICAM-1 in D group were significantly lower than those in DM. Conclusion Simvastatin has renal protective effect partly through reducing NF-κB and ICAM-1 expression.
