摘要
[目的]探讨大蒜油对胃癌SGC7901细胞增殖的抑制作用。[方法]①胃腺癌SGC7901细胞在体外常规培养,给予不同浓度大蒜油处理,观察细胞生长情况,用四唑盐(MTT)比色法检测细胞增殖,计算抑制率。②细胞给予大蒜油或转化生长因子α(TGF-α)处理,或同时给予大蒜油和TGF-α处理,用MTT比色法检测细胞增殖情况,计算抑制率。[结果]①胃癌SGC7901细胞常规培养,加入5%、10%、15%大蒜油作用72 h时,其细胞增殖明显受抑制,抑制率分别为24.5%、44.0%和54.8%。②加入2.5%低浓度大蒜油作用72 h,与2.5%血清培养比较,细胞增殖速度无明显变化;加入30μg/L TGF-α时,细胞增殖明显加快;当同时加入2.5%大蒜油和30μg/L TGF-α时,则TGF-α的促增殖作用明显受抑制,抑制率为18.1%。[结论]大蒜油能够抑制胃癌细胞增殖,并能抑制TGF-α刺激的胃癌细胞增殖。提示大蒜油可能对胃癌细胞的TGF-α自分泌、旁分泌促增殖环路具有抑制作用。这可能是大蒜油抗肿瘤细胞增殖的作用方式之一。
[Objective] To explore the inhibitive effect of garlic oil on the cell proliferation of gastric carcinoma.[Methods] (1)Human gastric vitro with routine method and treated by garlic adenocarcinoma SGC7901 cells were cultured in oil. Cell proliferation was detected with MTT method. Then the inhibitive rate was calculated. (2)Cells were treated by garlic oil or transforming growth factor alpha (TGF-α), and synchronously by garlic oil and TGF-α. And Cell proliferation was detected with MTT method. Then the inhibitive rate was calculated. [Results] (1)Cell proliferation was inhibited markedly by garlic oil. When the cells were treated by 5 %, 10 % and 15 % garlic oil for 72 h, the inhibition rate of the cell proliferation was 24. 5 %, 44. 0 % and 54. 8% respectively. (2)The cell proliferation speed in 2. 5 % garlic oil had not markedly difference with that in 2. 5 % calf serum after treated for 72 h. When the cells were treated by 30μg/L TGF-α, the cell proliferation accelerated markedly. When the cells were treated by 2. 5 % garlic oil and 30μg/L TGF-α, the cell proliferation stimulated by TGF-α was inhibited 18. 1 %. [Conclusion]Garlic oil can inhibit the cell proliferation of gastric carcinoma cultured not only in routine but also in the condition stimulated by TGF-α. One of its mechanisms maybe relate to its inhibitive effect on the autocrine and paracrine loops of TGF-α in gastric carcinoma cells.
