摘要
采用高效液相色谱法测定鸡蛋中的胆固醇含量,以C18(5μm,250mm×4.6mm)为固定相,乙腈-异丙醇(4∶1)为流动相,流速为0.5mL/min,检测波长为210nm,测定结果是,胆固醇浓度在0.1 ̄0.8g/L范围内与色谱峰面积呈良好的线性关系。结果表明,不同鸡蛋品种中胆固醇含量差异较大。该方法快速,灵敏,简便易行,无须皂化便可直接定量。
A reversed-phase high performance liquid chromatographic assay was applied for the rapid determination of cholesterol content in egg yolk. The C18 column (5 μ m, 250 mm × 4.6 mm) was selected and the mobile phase consisted of ace- tonitrile and 2-propanol (4 : 1) at a flow rate of 0.5 mL/min. The detection wavelength was 210 nm. A hnearity was obtained from 0.1 to 0.8 g/L of cholesterol with a good correlation coefficient. Omission of saponification has made this method more convenient, and can be used in yolk cholesterol assay.
