摘要
目的:观察补脾方药对脾虚模型大鼠壁细胞胃泌素受体及细胞内钙离子浓度的影响。方法:采用EDTA-Pronase酶法消化大鼠壁细胞,放射配基结合实验检测胃泌素受体,双波长荧光法测定壁细胞内钙离子浓度([Ca2+]i)变化。结果:模型组大鼠壁细胞胃泌素受体结合位点数显著降低(P<0.05),经补脾方药治疗后有所升高;同时模型组大鼠壁细胞内静息[Ca2+]i明显低于正常对照组(P<0.01),经胃泌素刺激后壁细胞内[Ca2+]i有所升高,且模型组与党参组升高幅度明显高于正常对照组(P<0.05),经治疗后此状态有所恢复,但差异无显著性。结论:脾虚证可能与胃泌素受体低表达相关,且壁细胞内钙离子处于低代谢状态,但对于胃泌素刺激的反应性增强,进而可能导致酸分泌敏感性相对增高,胃黏膜易损性增强,而补脾方药对其有一定的调节作用。
Objective :To observe effect of recipe or herbs of invigorating the spleen on gastrin receptor and intracellular calcium ion [Ca^2+ ]i level of parietal ceils in spleen deficiency model rats. Methods:The parietal cells of the rats were isolated by EDTA-Pronase method. Gastrin receptor was detected with radioligand binding assay and intracellular calcium ion level was determined with double wave-length fluorescence method. Results: The gastrin receptor binding sites of parietal cells in the model group were significantly decreased (P〈0.01) ,and they were increased after treatment by the herbs or recipe of invigorating the spleen. The intracelluler resting [Ca^2+ ]i of parietal cells in the model group was significantly lower than that in the normal group (P〈0.01), and it increased after stimulation of gastrin. The increasing amplitude in the model group and the Dangshen group was significantly higher than that in the normal group(P〈0. 05),and after treatment the condition was not significantly changed. Conclusion:Spleen deficiency syndrome is possibly related with low expression of gastrin receptor,and calcium ion of parietal cells is at low level. But the reactivity to gastrin stimulation was strengthened,possibly further inducing relative increase of sensitivity of acid secretion and injury of gastric mucosa, on which the medical herbs or recipe of invigorating the spleen have regulative action.
