补肾法对去卵巢大鼠骨髓细胞OPG、RANKL和TNF-α基因表达的影响 被引量:11

摘要 目的观察补肾中药对大鼠去卵巢后骨髓细胞表达核因子κB受体活化子配体(RANKL)、核因子κB受体活化子(RANK)、护骨素(OPG)和肿瘤坏死因子α(TNFα)基因的影响。方法健康3月龄雌性SD大鼠72只,其中24只为假手术对照组,48只去卵巢后随机分为去卵巢组和中药组,每组24只。分别于去卵巢后2、4、6、12周每组各取6只大鼠骨髓细胞提取RNA,RTPCR半定量分析其表达。结果与对照组比较,去卵巢后2周,去卵巢组骨髓细胞TNFα和RANKL的mRNA表达显著升高(P<0.05~0.01),第4~6周,上述改变达高峰,至第12周TNFα仍呈有意义增高;而去卵巢组OPG表达在第2~6周则明显降低,2~4周为最低点。中药组TNFα和RANKL表达低于去卵巢组(P<0.05,P<0.01),而OPG表达明显高于去卵巢组。但是中药治疗未能使以上基因表达完全恢复正常。各组全程未见RANK基因表达水平有明显变化。结论补肾中药在一定程度上抑制去卵巢大鼠骨髓细胞过度表达TNFα和RANKL,同时增加OPG的表达。 Objective To observe the effect of the Chinese herbs (HB) for invigorating the kidney on gene expression of receptor activator of NF-κB ligand (RANKL), receptor activator of NF-κB (RANK), osteopretegerin (OPG) and tumor necrosis factor-α(TNF-α) in myeloid cells of ovariectomized rats. Methods Seventy-two healthy female SD rats, 3 months in age, were randomly divided into three groups: 24 in the sham-operated control group (A), 24 in the ovariectomized group(B) and 24 in the HB treated group after ovariectomy (C). Bone marrow cells of 6 rats from each group were respectively collected at different times (2^nd ,4^th ,6^th and 12^th weeks after operation) and the total RNA was extracted by TRIZOL and analyzed with semi-quantitative RT-PCR. Results Compared with group A, significantly increased expression of RANKL and TNF-α in Group C was found since the 2^nd week ( P 〈 0.05 or P 〈 0.01 ) with the peak value at the 4^th to 6^th week. The increased expression of TNF-α remained to the 12^th week. However, the OPG gene expression decreased significantly in group C at week 2 ~ 6 with the lowest level at week 2 ~ 4. The expression of RANKL and TNF-α in group C was significantly lower than that in group B (P 〈 0.05 or P 〈 0.01) with a higher OPG level. But HB did not completely restore the changes of the above gene expressions. No significant change of RANK gene expression was revealed in this experiment in any of the three groups. Concluslon HB could partly inhibit the over-expression of TNF-α and RANKL genes and promote OPG gene expression in myeloid cells in ovariectomized rats.
