缬沙坦和美托洛尔对心肌梗死后大鼠钾通道Kv9.X基因表达的不同影响

摘要 目的探讨肾上腺素能β1受体阻滞剂和肾素-血管紧张素Ⅱ1型受体阻滞剂对心肌梗死大鼠钾通道Kv9.X基因表达的影响。方法选用SD雄性大鼠建立心肌梗死模型,取存活大鼠随机分为心肌梗死组、缬沙坦组、美托洛尔组,同时建立假手术组。4组大鼠均再分为7d和30d两亚组。缬沙坦组和美托洛尔组大鼠分别于术后4h^12h给予缬沙坦(30mg/kg)和美托洛尔(8mg/kg)灌胃至术后相应时间,术后第7天和第30天留取大鼠心肌组织,提取心肌总RNA和检测Kv9.1、Kv9.2、Kv9.3mRNA。结果美托洛尔组与心肌梗死组比较,30d钾通道Kv9.XmRNA表达明显升高,但与缬沙坦组比较降低、与假手术组比较无论7d组还是30d组均明显降低。缬沙坦组与假手术组比较,无论是7d组还是30d组均无统计学意义,与心肌梗死组比较显著升高。结论美托洛尔与缬沙坦均可提高心肌梗死后大鼠心肌Kv9.1、Kv9.2、Kv9.3的表达,缬沙坦作用更为显著。
