摘要
目的探讨迷迭香酸(rosmarinicacid,RosA)对谷氨酸(glutamate,Glu)诱导PC12细胞损伤的作用和机制。方法以谷氨酸损伤PC12细胞为模型,用甲氮甲唑蓝(MTT)法检测细胞存活状况,碘化丙啶(propidiumiodide,PI)染色流式细胞术检测细胞凋亡,RTPCR检测bclxl和bax基因的表达。结果用不同剂量谷氨酸(1、5、10、12.5、15、20mmol·L-1)孵育细胞24h后,细胞活性随药物浓度的增加依次下降,RosA(100μmol·L-1)预处理1h后可以改善PC12细胞的存活率,存活率由55.65%±5.07%增加到63.74%±2.04%;使PC12细胞的凋亡率由21.11%降至3.24%,并且使bclxl基因表达上调和bax基因表达下调。结论迷迭香酸有抗谷氨酸诱导PC12细胞凋亡的作用,其机制可能与调节bclxl和bax基因的表达有关。
Aim To investigate the effect and mechanism of Rosmarinic acid ( Ros A )against PC12 cell injury induced by glutamate. Methods With the model of PC12 cell injury induced by glutamate,the cell viability was observed by MTY assay; the cell apoptosis was detected by flow cytometric analysis (FCM);the expression of bcl-xl and bax gene were determined by RT-PCR. Results After PC12 cell was incubated with different concentration glutamate ( 1,5,10,12. 5, 15,20 mmol · L^-1 ) for 24 hour, the cell survival of PC12 cell was reduced in a concentration-dependent manner. However, after one hour's pretreatment of Ros A( 100μmol · L^-1) ,the cell survival of PC12 cell was promoted, from 55.65% ± 5.07% to 63.74% ± 2. 04% ;the apoptotic rate of PC12 cell was decreased, from 21.11% to 3.24% ;and the expression of bcl-xl gene was increased and the expression of bax gene was decreased. Conclusion Ros A can resist the injury of PC12 induced by glumatame,and the mechanism of antiapoptosis may be related to regulation of the expression of bcl-xl and bax genes.
