兔髁突软骨细胞与羊膜间生物相容性研究 被引量:3

摘要 目的:研究羊膜作为颞下颌关节组织工程支架材料应用的的可行性。方法:采用Ⅱ型胶原酶消化解离兔髁突软骨中的纤维软骨细胞,取经TGF-β1扩增的第二代细胞与羊膜复合培养,进行MTT、光镜、扫描电镜观察。结果:髁突细胞于羊膜上牢固黏附,第3天开始增殖,3周时已有大量细胞生长于羊膜上。结论:羊膜表现出良好的吸水性、细胞吸附性和体外生物相容性,髁突细胞可在其上黏附、增殖,羊膜适于作为髁突组织工程支架材料。 Objective: To investigate the possibility of amnion as cell scaffold in tissue engineering of temporomandibular joint. Methods: Rabbit mandibular condylar fibrocartilage was digested by collagenase, and cells of the second generation proliferated by TGF - β1 were cultured onto amnion. Observation was made with MTT, light microscope, and electronic scanning microscopy. Results: Fibrocartilage cells attached to amnion closely and began to proliferate on the 3rd day. In 3 weeks after the transplantation, a lot of cells grew well on amnion. Conclusion: Amnion has good water adsorptivity and biocompatibility with fibrocartilage cells. It can be used as scaffold for tissue engineering of temporomandbular joint.
